Objective: To survey the changes of Toll-like receptor 4(TLR4)/ activator protein AP-1 signaling pathway and tumor necrosis factor(TNF-?),interleukin-6(IL-6)in the H9c2 myocyte hypertrophy induced by isoprenaline for studying the intervention effects and mechanisms involved of Fritillaria thunbergii extractMethods:(1)MTT assay was used to detect the effects of Iso,Fritillaria thunbergii extract and captopril on the survival rate of H9c2 cardiomyocytes,and to determine their safe concentration range.(2)In the concentration range of Iso to H9c2 cardiomyocytes,a concentration gradient was established,and the best concentration was selected.(3)Cell morphology was observed by crystal violet staining and inverted fluorescence microscope.Image-pro plus 6.0 software was used to measure the area of cardiomyocytes in each treatment group.(4)to determine the total protein content of each group by Diacetic acid(BCA)method(5)to detect the concentration of TNF-? and IL-6 in extracellular fluid by Enzyme-linked immunosorbent assay(ELISA)(6)Western blot was used to detect the protein expression of TLR4,AP-1 and ANP.Results:(1)The safe concentration of Iso to H9c2 cardiomyocytes was within 30?mol / L;the safe concentration of Fritillaria thunbergii extract to H9c2 cardiomyocytes was in the range of 2000?g / mL;Captopril on H9c2 cardiomyocytes The safe concentration range is within 0.2 m mol / L.(2)Iso's best molding concentration of 20?mol / L.(P <0.001),and the surface area of H9c2 cardiomyocytes in the model group was significantly higher than that of the other groups,The surface area of H9c2 cardiomyocytes in the high dose group was significantly lower than that of the other groups(P <0.001)which has a protective effect for cardiomyocyte hypertrophy.(4)The total protein content of the model group was significantly higher than that of the other groups(P <0.001).The total protein content of the positive control group was close to that of the normal group.(5)The concentration of TNF-? and IL-6 in the extracellular fluid of the model group was significantly higher than that of the model group(P <0.001).(6)The protein expression of TLR4,AP-1 and ANP in the model group was up-regulated,and the expression of these proteins was down-regulated in high dose group and positive control group.Conclusion: Fritillaria thunbergii extract may have protective effects on the H9c2 cardiomyocyte hypertrophy induced by Iso.Which mechanisms may be related to decrease the release of TNF-? and IL-6,inhibite the TLR4 / AP-1 signaling pathway,and to reduce the inflammation of myocardial hypertrophy.
