Background Coronary artery disease(CAD)is the leading cause of death in the global cause.The role of genetic factors in coronary heart disease is of concern.Platelet-derived growth factor D gene(PDGFD)is a member of the platelet-derived growth factor family of proteins,it is involved in the body 's development and physiological processes,but also with the tumor,fibrous diseases and atherosclerosis Hardening related.Studies have found that PDGFD transcripts are highly expressed in the heart.The product promotes atherosclerosis by affecting the activity of matrix metalloproteinases,monocyte migration,and inhibition of smooth muscle cell gene expression.Genome-wide association study of the results of the survey of Korean people show that PDGFD gene rs974819 polymorphism and coronary heart disease there is a significant relationship,due to differences in different racial genes,there is no Chinese Han Chinese related information.Objective To investigate whether PDGFD gene polymorphism is associated with coronary heart disease in Yubei Han population;and to explore its association with coronary heart disease risk factors and severity of lesion by genotyping,and to provide more basis for early intervention and risk assessment of coronary heart disease in Han population.Methods1.Genomic DNA extraction All subjects were extracted 5ml venous blood samples,EDTA-K2 anticoagulation,using QIAamp DNA Blood Mini Kit kit to extract peripheral blood DNA,all samples were detected using Nano Drop-2000 trace nucleic acid protein detector,80 ? save standby.2.The target gene was designed and the target gene fragment was amplified by PCR.The target gene was amplified by electrophoresis.3.Genotyping The Big Dye v3.1 kit was successfully sequenced on the ABI 3730 XL sequencer at GENEWIZ,Inc(Beijing).4.Statistical analysis of SPSS 21.0 statistical software.Results1.There was no significant difference in age and gender between the two groups(P>0.05),but the proportion of smoking,diabetes mellitus,hypertension and hyperlipidemia in coronary heart disease group was significantly higher than that in control group(P<0.05)At the same time,high density lipoprotein cholesterol in coronary heart disease group was lower than that in control group(P <0.05).2.There were three genotypes of AA,AG and GG in the rs974819 polymorphism of PDGFD gene.3.The frequencies of alleles in the coronary heart disease group were 60.9% and39.1%,respectively,and 64.0% and 36.0% in the control group,respectively.There was no significant difference in allele frequency distribution between the two groups(?2=0.63,P>0.05).AA,AG and GG genotypes were 35.8%,50.3% and 13.9% in the coronary heart disease group,42.6%,43.6% and 13.8% in the control group,respectively.There was no significant difference in genotype frequencies between the two groups(?2=1.37,P>0.05).The allele frequencies of A allele in Yubei Han population were 0.609 and 0.640,respectively,which were higher than that of European population A allele frequency 0.410,PDGFD gene locus A,G allele and AA,AG,GG genotype There was no significant difference in the allele and genotype frequencies between the coronary heart disease group and the control group(P> 0.05)4.The Gensini Score scores of AA,AG and GG genotype in patients with coronary heart disease were 53.47,42.28 and 34.92,respectively,The difference was statistically significant(P <0.05).Conclusion1.The rs974819 polymorphism of PDGFD gene may not be associated with genetic susceptibility to coronary heart disease in Yubei Han population.2.There is relation between PDGFD gene mutations homozygous and coronary heart disease severity,and the patients of coronary heart disease carrying PDGFD gene mutation homozygous AA genotype stenosis heavier.
