Background ObjectiveTo explore the expression of apoptosis-stimulating protein of p53(ASPP)family members in invasive breast cancers,and their relationships with clinical-pathological characteristics and prognosis of patients.MethodsThe invasive breast cancer samples were collected from Feb.2005 to Apr.2010 in 150 th Hospital.There were 1 man and 154 women in total of 155 patients aged 25~84,and the survival time was 2~108 months.The expression of ASPP family members(ASPP1,ASPP2,iASPP),ER,PR,HER-2,p53 and ki-67 in breast cancers were detected by immunohistochemistry,the expression of HER-2 protein was observed by fluoresce in situ hybridization,and the relationship between ASPP family members expression and clinic pathological characteristics,prognosis of breast cancer was analyzed by SPSS 13.0 software.Results1.ASPP1 was observed in the cytoplasm and nuclei of tumor cells,ASPP2 was found on the nuclei,whereas iASPP was found in the cell cytoplasm.The results showed that there were 21 cases ASPP1 expression(21/155,13.55%),151 cases ASPP2 positive(151/155,97.42%),95 cases iASPP expression(95/155,61.29%).2.There were difference between ASPP1 expression and pTNM stage(P<0.01),between ASPP2 expression and histological grade(P<0.05),and there were no difference between iASPP expression and clinic pathological characteristics in breast cancers(P>0.05).3.There were 55 cases of p53 expression in 155 cases of breast cancers.There were no difference between ASPP family members and p53 expression.4.There were 44 cases Luminal A subtype breast cancers,66 cases Luminal B subtype,18 cases basal-like subtype and 27 cases HER-2 over-expression subtype in all of 155 patients,there were no relationship between ASPP expression and different subtype of breast cancers.5.The 3-years survival rate was 84.52(131/155)and 5-years survival rate was 79.35%(123/155)in 155 cases.The difference was observed between ASPP1 positive group and negative group in 3-years,5-years survival rate(x2=4.49,P<0.05;x2=3.85,P<0.05).There were no difference between ASPP2 positive cases and negative cases in 3-years,5-years survival rate(x2=0.28,P>0.05 and x2=0.04,P>0.05).And the difference was observed between i ASPP positive group and negative group in 3-years survival rate(x2=3.02,P>0.05),not in 5-years survival rate(x2=1.25,P>0.05).Conclusions1.The expression model of ASPP family members in breast cancers were different,ASPP1 expression can be used to assess the pTNM stage of breast cancer,ASPP2 expression can be used to judge histological grade.2.There were difference between ASPP1 positive cases and negative cases in 3-years and 5-years survival time,the results indicated that ASPP1 can be used to predict the prognosis of patients.3.The over-expression of ASPP2 was observed in invasive breast cancers,ASPP2 maybe a candidate molecular target of breast cancer.
