Neurotrophin(NT)was first discovered in the nervous system and involved in the growth and differentiation of nerve cells.NT was composed of nerve growth factor(NGF),brain-derived neurotrophic factor(BDNF)NT-3,NT-4/5 and NT-6 and NT-7 derived from mammals.There are currently two types of NT receptors,namely the low affinity receptor p75 neurotrophic factor receptor and high affinity Tyrosine kinase receptor(Trk)and Trk can be divided into three types,TrkA,TrkB and TrkC,which selectively bind to different NT,and the two receptors are often in the same neuron.At the same time,the expression of common regulation of neuronal response to NT.In recent years,more and more studies have shown that neurotrophic factors and their receptors are associated with tumor growth,including the promotion of tumor cell proliferation,tumor cell survival and infiltration,studies of breast cancer,ovarian cancer,prostate cancer,colorectal cancer have confirmed the above view,and neurotrophic factor and its receptor in cervical cancer in the study of little,the existing research is not yet comprehensive,the number of cases included in are limited.So this study is mainly on neurotrophic factor and its receptors In cervical squamous cell carcinoma from the view of qualitative and quantitative research,and analysis of the relationship between the different observation indicators,and further deduced its role in the development and progression of cervical squamous cell carcinoma,and provided the direction for the follow study on the mechanism of cervical cancerChapter 1 Expression investigate the qualitative and quantitative status of neurotrophic factor and its receptor in normal cervical tissuesMethods:Immunohistochemistry,biology and visualizationResults:(1)NGF was expressed only in the basal layer of normal cervical epithelium,which was linearly distributed and showed an average optical density of 0.0347±0.0567.BDNF expression was evenly distributed in the normal cervical epithelium.The expression of P75 in the cervical epithelium was decreased from the basal cell layer to the keratinized layer,which was the expression of the cell membrane and cytoplasm,and the average optical density was 0.0482±0.0239;TrkA and TrkB were evenly distributed in the normal cervical epithelial cells,showing the co-expression of nucleus and cytoplasm.The average optical carcinomaMethods:Immunohistochemistry,immunofluorescence and biology were investigated Results:(1)NGF was mainly located in the nuclei of cancer cells,the positive rate was 92.4%,the difference between the expression level and the expression level in normal cervical epithelial cells was not statistically significant(0.0321±0.0254:0.0347±0.0567,t=(P<0.05).BDNF and P75 were mainly located in the cell membrane,the positive rate was 100%,compared with normal cervical epithelial cells,the former expression of low(0.1409±0.0550:0.2560±0.0992,t =-4.421,(0.1072±0.0694:0.0482±0.0239,t=6.797,P<0.05).TrkA was mainly located in the cytoplasm,the positive expression rate was 96.8%,the TrkB was mainly expressed in the membrane,(0.0844±0.0658:0.0506±0.0390,t=6.797,P<0.05),and the expression level was low(0.0421±0.0334:0.1199±0.0390),which was higher than that in normal cervical epithelial cells(3)In the squamous cell carcinoma of the cervix,we can see that the P75 positive nerve(P<0.05),(2)the expression of the neurotrophic factor and its specific receptor in the cervical squamous cell carcinoma Fiber,the positive rate was 50.8%(63/124),TrkA and TrkB visible table(P<0.05).(4)The expression of BDNF and TrkB,P75 and TrkA,TrkA and TrkB in cervical squamous cell carcinoma were significantly higher than those in the control group(P<0.05),and the expression of BDNF and TrkA,TrkA and TrkB in cervical squamous cell carcinoma(R=0.452,P<0.05),and there was no correlation between the other neurotrophic factors and the expression levels of neurotrophic factors(r=0.452,P<0.05).Conclusion:(1)The changes of neurotrophic factor and its receptor expression were related to the occurrence of cervical squamous cell carcinoma,and the expression of each factor was related to the relationship between the expression of each factor in the tumor cells and promote the growth of tumor cells(2)The correlation structure of neurotrophic factor receptor positive in stroma interstitium,indicating that neurotrophic factor receptor may have an effect on tumor growth microenvironmentChapter two Expression of neurotrophic factor and its relationship with tumor staging in cervical squamous cell carcinomaObjective:To study the relationship between qualitative and quantitative expression of neurotrophic factor and its receptor and stagingMethods:The Wilcoxon rank sum test and the single factor analysis of variance were used to analyze the clinical and pathological indexes and experimental results of spearman rank correlation,two independent samples.(X~2=22.841,P<0.05),(X~2=23.442,P<0.05),and the other factors were significantly higher than those in the control group(P<0.05),and the expression of BDNF and P75(F=5.303,P<0.05).The expression level of NGF in ?BA-?B was the highest(P<0.05),and the expression level of NGF was the highest(0.0571±0.0281),and ?B1-?A2 expression and staging showed an upward trend,the other factors and receptor expression in the staging between the no difference;(3)P75 receptor and TrkA receptor expression ratio There was a negative correlation between the two groups(r=-0.257,P<0.05).Conclusion:The expression level of NGF,the ratio of P75 to TrkA receptor expression,the expression of BDNF and P75 were correlated with the
