Objective: To investigate pathological morphology of liver,changes and significances of TLR-4 and MMP-7 in Rats model infected with Clonorchis sinensis.Methods: 50 female SD rats were divided into 2 groups.The experimental group rats were orally infected with 200 metacercariaes of Clonorchis sinensis,and the normal rats were give same amount of saline as a control group.Following identification of infection,all rats were re-divided into three groups: uninfected group was given 25 mg/L DEN water(group A),Clonorchis sinensis infected jointed 25 mg/L DEN water group(group B),Clonorchis sinensis infected jointed ordinary drinking water(group C).2-3 rats in every group were random sacrificed following identification of infection in 0 W,2 W,5 W,9 W,13 W,17 W,respectively.Sacrificed rats were used to get serum and take the left liver leaves near liver door department 2*2*2 mm tissue,fixed with 10% neutral formaldehyde solution to make general HE dyeing.TLR-4,MMP-7,IL-6 and TNF-? in serum were detected by ELISA.MMP-7 in liver tissue of Group B was detected by Western Blot.Results: Contents of IL-6 and TNF-? in infected group and uninfected group showed statistically significant differences(t=2.623,P<0.01)and statistically significant differences among the three groups(F=4.38,P<0.05),additionally,statistics differences in group B were showed(F=5.22,P<0.05).Changes of MMP-7 content had positive correlation trend with IL-6 and TNF-? at the coefficients of 0.832 and 0.782,respectively.HE dyeing showed biliary dilatation,wall thickening obvious swelling of liver cells,liver lobule structure damaged,biliary epithelial cells arranged irregular edema,necrosis,Hepatobiliary tube blocked seriously,while caused bile duct meeting zone became widely,Significant expansion of bile deduct,small increased in bile duct,biliary interstitial fibrosis obvious.Conclusions: Expression level of TLR-4 in the progression of rats infected with Clonorchis sinensis increased at first then reduced,and then increased,suggested TLR-4 played an important role in the connectivity of innate immunity and humoral immunity.MMP-7 was produced in group infected with Clonorchis sinensis and expression level was increased during the infection process,and showed positive correlation trend with inflammatory factors of IL-6 and TNF-?,indicated the association of final development of the bile duct lesions.Clonorchis sinensis infection combined with 25 mg/L DEN accelerated the progression of hepatic duct deterioration,exhibited more severe symptoms compared with the effect of simple Clonorchis sinensis infection and the characteristics of typical bile duct cancer had not appeared in 17 weeks after infection,suggested if the model of inducing bile duct cancer to be created should prolong the time of infection or increase DEN concentration.TLR-4 and MMP-7 might be used as indicators for monitoring the occurrence and development of cholangiocarcinoma,while the contents of both needed to be studied further.
