Objective: Studied on the expression of HOXA5(Homeobox A5),E-cadherin and ?–catenin in CSCC(cervical squamous cell carcinoma,CSCC)and chronic cervicitis.Next,we assessed the correlation between this three factors referring above and the clinicopathological data in CSCC patients.To further investigated the role of HOXA5 in the occurrence and development of cervical cancer.Methods: Cervical carcinoma tissue specimens were obtained from 120 patients with histologically confirmed cervical squamous cell carcinoma who underwent surgical resection at the affiliated Hospital of Guilin Medical University between 2007 and 2014,and their mean age was 48.7 years.In addition,30 cases of chronic cervicitis were used as control.Detection of HOXA5,E-cadherin and ?-catenin at the protein level by immunohistochemistry in CSCC and chronic cervicitis,and 54 patientsamong these patients were analyzed about prognosis.At the same time,RT-PCR was used to detect HOXA5 mRNA expression in cervical cancer cells Hela,Si Ha,CaSki and C-33 A.Using statistical software SPSS18.0to analyze experimental results above,Pearson's Chi-Square(?2)and Fisher's exact test was used to assess the differential expression of HOXA5,E-cadherin and ?-catenin in CSCC and chronic cervicitis.?2 test was used for correlation analysis.The overall survival time of CSCC patients was evaluated by using the Kaplan-Meier method.The statistically significant level is p ? 0.05.Result:(1)The positive expression rate of HOXA5,E-cadherin and ?-catenin in CSCC was36.7%(44/120),40.8%(49/120),and 34.2%(41/120)respectively,which is different from the positive expression rate of chronic cervicitis.The differences were statistically significant(p ? 0.05)respectively.(2)The decrease of HOXA5,E-cadherin and ?-catenin expression was correlated with tumer differentiation degree(p?0.05)respectively,but not with age,myometrial invasion and lymph node metastasis(p>0.05).(3)There was positively correlated with expression of HOXA5,E-cadherin and?-catenin in CSCC(p value was 0.020,0,000 respectively).(4)Patients with higher expression of HOXA5 had much more favorable prognosis than those with lower expression(P =0.044),but E-cadherin and?-catenin had no feature(P > 0.05).(5)In Hela,SiHa,CaSki and C-33 A four cervical cancer cell lines,the expression of HOXA5 mRNA in SiHaand C-33 A was significantly lower than that of Hela and CaSki.Conclusions:(1)The expression of HOXA5,E-cadherin and ?-catenin in CSCC was significantly lower than the control group.The expression of HOXA5,E-cadherin and ?-catenin in CSCC was correlated with the tumor differentiation degree.HOXA5 expression was positive correlation with E-cadherin and ?-catenin expression in patients.It is suggested that HOXA5 may be related to the occurrence and development of cervical cancer.(2)The high expression of HOXA5 predicted well prognosis of patients after operation.(3)The expression of HOXA5 mRNA in cervical cancer cell lines SiHa and C-33 A was significantly lower than that of Hela and CaSki.(4)The changes of HOXA5,E-cadherin and ?-catenin expression levels in CSCC play an important role in the occurrence,development and prognosis of disease.
