Objective:To study the inhibitory effect of resveratrol on SMMC-7721 cells and to study the mechanism of resveratrol on hepatocarcinoma cells.Methods: different concentrations of Resveratrol was administered to SMMC-7721 cells,The effect of resveratrol on the proliferation of SMMC-7721 cells was detected by growth curve and clone formation assay,resveratrol detected cell cycle of SMMC-7721 cells by Flow Cytometry,western blot was used to detect the changes of m-TOR protein and phosphorylation level of SMMC-7721 cells by resveratrol.Results: Resveratrol intervened in SMMC-7721 cells for 48 hours,in a certain concentration range,the effects of resveratrol on the inhibition of SMMC-7721 cells in a dose-dependent manner,the inhibition rates were 9.55% ± 7.33%,8.96% ± 6.97%,12.61% ± 10.52%,17.18% ± 10.42%,16.11% ± 1.51%,20.12% ± 7.73%,21.56% ± 6.62%,32.23% 3.36%,55.73% ± 0.37%,56.53% ± 0.11%,the differences had statistical significance(F=21.067,P<0.01).In the range of drug concentration,resveratrol inhibited SMMC-7721 cells in a time-dependent relationship,the differences had no statistical significance after treated 2 hours,but had statistical significance after treated 2,4 and 6 days(F=11.860,257.439,181.780,P<0.01).and the 4 days Inhibition rate were 6.76%±5.30% ? 12.30%±7.01% ? 26.59%±3.66% ? 42.97%±4.52% ?61.65%±2.35%?67.88%±1.36%.Colony formation test showed that resveratrol could inhibit the SMMC-7721 cells proliferation significantly,and colony inhibition rates increased with the concentration of drugs.With different doses groups treated SMMC-7721 cells 4 days,the colony inhibition rates were 6.76%±5.30% ? 12.30%±7.01% ? 26.59%±3.66% ? 42.97%±4.52% ?61.65%±2.35% ? 67.88%±1.36%,the differences had statistical significance(F=195.978,P<0.01).Resveratrol block SMMC-7721 cells in G0 / G1 phase after treated 48 hours,cell ratio of G0 / G1 phase were 60.22%±105.22% ? 60.39%±64.49% in control groups,61.14%±36.29% ?60.47%±35.7% ? 45.85%±109.04% ? 48.15%±138.02% ? 90.13%±95.41% ?76.01%±35.23% in resveratrol groups,the dffect of R12.5?R25?R50?mol·L-1 groups in dose-dependence(F=1894.538,P<0.01).Westren blot assay showed that m-TOR expression had no difference Compared with the control,but resveratrol Reduced m-TOR phosphorylation levels.Conclusion: Resveratrol inhibited human SMMC-7721 cell proliferation,arrested the cell cycle at G0 / G1 phase,the inhibition mechanism may be related to activation of m-TOR phosphorylation levels by resveratrol.
