Objective:Anti-AD effect of 5-substituted-1-azaanthraquinone derivatives A7 on AD cell model and its possible mechanism are studied in this paper,which provides preliminary study basis for finding out a kind of new drug for delaying AD occurrence and development.Method: Managing APPsw cell with the5-Replacing-1-Nitrogen Impurity Anthracene Ketone Derivative solution with the concentration being 0.01,0.1,1,10,?M,respectively,and detecting the cytotoxicity of compound A7 with MTT experiment;detecting the A?1-42 secretion level of APPsw cell with ELISA experiment;analyzing the effect of compound A7 on A? precursor protein gene APP expression with western blot technology.AChE measure is detected through the reaction principle of choline and DTNB.The effect of APPsw cell after oxidative damage is evaluated with fluorescence detection method and flow cytometry.Results: Through this study,the anti-AD activity and capacity of 10 kinds of composite 5-replace-1-nitrogen impurity anthracene ketone derivative are evaluated and selected.Results showed that in this 10 kinds of compound,A7 has the most significant effect inreducing the A?1-42 secretion level of APPsw cell.Moreover,it is also found that compound A7 can lower the APP protein expression.It is further observed that compound A7 reduced AChE activity of APPsw cell.The compound A7 inhibited hydrogen peroxide induced damage in APPsw cell.Conclusion: The study data showed that 5-replace-1-nitrogen impurity anthracene ketone derivative A7 is beneficial to reduce AD morbidity,which lays the study foundation for researching and developing a kind of newtype drug to delay the occurrence and development of AD.
