Objective:Explore the effect of Vitamin D on the expression level of the surface molecules CD83,CD86 of pulmonary dendritic cells and secretion level of anti-inflammatory factor IL–10 and proinflammatory factor IL-12 in septic rats.Methods:Divide 24 juvenile male(SD)rats according to random number table into normal control group,sepsis group and vitamin D(VitD)intervention group.Intraperitoneal injection of lipopolysaccharide(LPS 5mg/kg)preparation in the rat model of sepsis,VitD3(1000U/kg)is injected intraperitoneally with 48 h,24h and 1h before the sepsis model being established in VitD group.12 hours after sepsis model preparation,we will collect blood and lung tissue of rats of all groups.Using flow cytometry instrument to detect lung DC surface molecules CD83 and CD86(expression);Using enzyme-linked immunosorbent assay(ELISA)to check the content of of proinflammatory factor(IL-12)and anti-inflammatory factor(IL-10)in lung tissue;Using the ELISA method to check the levels of each rat serum 25(OH)D.Results:For juvenile rats,after injection of LPS,the expression of lung DC surface molecule CD86 and CD83 go up,compared with normal control group,expression rate is obviously higher,and significantly different(P < 0.05).But the expression of VitD group lung DC surface molecule CD86 and CD83 reduces,compared with LPS group,which has significant difference statistically(P<0.05);The content of IL-12 of homogenate supernatant of lung tissue in LPS group is higher,while the level of IL-12 lower.Both two kinds of inflammatory factors mentioned above have statistically difference,compared with normal control group.Secretion level of proinflammatory factor IL-12 in VitD group’s lung tissue is lower than that in LPS group,but anti-inflammatory factor IL-10 level is a significantly higher than that in LPS group.And,both have statistically difference.In three groups,The level of serum 25(OH)D in LPS group is the lowest;VitD group’s serum level is higher than LPS group,which is significantly different(P < 0.05).But the two groups were significantly lower than normal control group.Lung tissue pathology detection shows that LPS group’s lung tissue pathology change significantly.Whereas the change of VitD group’s lung tissue pathology is smaller than LPS group’s.Conclusion:1.Succeeding in preparing animal models of sepsis.2.In septic juvenile rats,the expression of lung DC surface molecule CD86 and CD83 goup,Secretion level of proinflammatory factor IL-12 of inflammatory cytokines increasing,IL-10 level decreasing,which means the activation of DCs & the secretion of inflammatory cytokines get involved in the occurrence and development of sepsis.3.VitD can inhibit the expression of lung DCs surface molecular CD83,CD86 of septic juvenile rats;can promote the secretion anti-inflammatory factor of IL – 10;can keep down the release of proinflammatory factor IL – 12 and reduces the body’s excessive reaction to inflammation.
