The Expression And Clinical Significances Of TRIM23 In Patients With Gastric Carcinoma

Objective: To explore the clinical significances and expression of TRIM23 in gastric carcinoma and discuss the relationship between TRIM23 and Hippo pathway.Methods: We collected 65 cases of tissue specimens of gastric carcinoma patients who received surgical operation in the first affiliated hospital of Nanchang university from 2009 to 2011.At the same time,we also obtain the 20 cases of pericarcinomatous tissue from these patients as control.Using immunohistochemical(IHC)to analysis the expression of TRIM23,the Pearson's ?2 test was used to evaluate the relevance between TRIM23 expression and clinicopathological parameters,and the log-rank test was applied to assess differences of overall survival(OS)between two groups(low TRIM23 expression vs.high TRIM23 expression).We use western blot to detect the expression of TRIM23 protein in gastric cancer cell lines and tissue.And Quantitative Real-time PCR(q RT-PCR)is used to detect the level of TRIM23 message RNA(m RNA)in gastric cancer tissue.Finally we discussed the relationship between TRIM23 and Hippo pathway via looking up databases on c Bio Portal.Results: 1)Results of western bolt indicated that TRIM23 protein overexpress in gastric cancer cell lines and tissues.2)q RT-PCR showed that TRIM23 m RNA is upregulated in gastric cancer tissues(p<0.05).3)Results of IHC confirmed that the expression of TRIM23 protein is significantly higher in gastric cancer tissues than in pericarcinomatous tissue(p=0.006).And the level of TRIM23 expression is closely related to TNM-staging(p < 0.001),depth of invasion(p=0.007),lymphatic metastasis(p=0.018),size of tumor(p=0.011)and nerve invasion(p=0.038).The overall survival of group with high TRIM23 expression is shorter than the group with low expression(p=0.025).4)The databases on c Bio Portal suggested that there are not sufficient evidences to support the relevance between TRIM23 and Hippo pathway.Conclusion: We find the overexpression of TRIM23 in gastric cancer which may be related with poor prognosis.So,TRIM23 may be regarded as an important and new mark in the diagnosis of gastric cancer,and might becomes a new therapeutic target in the future.
