Objectives:Hepatocelluar carcinoma is one of the most common malignant tumor in the worldwide.Recently,with the progress and development of liver cancer diagnosis and treatment technology,the percentage of surgical resection of liver cancer has greatly increased.However,patients are always faced with metastatic recurrence postoperation and 5-years survival rate is low.Phospholipid is the most widely distributed organism biological active substances.It is closely related to cancer pathological process.Therefore,it is of importance to look for new biomarkers to predict the metastatic recurrence of hepatocelluar carcinoma.Methods:1.Collected 40 patients with hepatocelluar carcinoma patients clinical specimens and made mass spectrum detection with carcinoma tissues and normal tissues according to the experiment criterion.All specimens have been proven by pathology H-E staining.2.Recurrent cancer was caused by circulating tumor cells and the human body specimen gets more difficult.The mice model of hepatocelluar carcinoma was established based on healthy mice.The mice model of situ with 20 cases and metastasis/recurrence model with 20 cases.After successing the model,specimens collection began according to the criterion.All specimens have proven by pathology H-E staining.3.Collected specimens were divided into control group and experimental group,control group as adjacent tissues and the experimental group as the carcinoma tissues.Use mass spectrometry to detect phospholipids abundance between two groups.The experimental group were layered,namely,primary tumors and metastases group and recurrence group,using mass spectrometry detection technology each subgroup phospholipids abundance.4.Principal componentanalysis with the mass spectrum data,to determine the various types mass-to-charge ratio of group differences in phospholipid,and identified the material by the secondary mass spectrometry.Result:1.Phospholipids expression between human cancer tissues and adjacent tissues are different.The main different ions were m/z 757,m/z 783,m/z 799,m/z 820.2.The main different ions of primary cancer tissues and metastasis/recurrence tissues were m/z 774,m/z 783,m/z 799,m/z 821.3.The main different ions of tissues were m/z 783,m/z 799.4.The different phospholipids wre sphingomyelin and phosphatidylglycerol.They were accociated with the size and metastasis of tumor(P<0.05).They were not accociated with age,gender,cirrhosis of the liver and AFP(P<0.05).Conclusion:1.In hepatocelluar carcinoma,normal liver tissues and the cancerous tissue,primary cancer tissues and cancer metastasis and recurrence of phospholipids expression have differences.2.The identified specific differences phospholipids are phosphatidylglycerol and sphingomyelin.3.The specific phospholipids are associated with clinical outcomes and can be used as a diagnostic potential biomarkers of liver cancer.
