Objective:In order to research the influences of ISG15 on cell proliferation,migration,invasion,apoptosis and cycle of Hep3 B cells by interfering with the expression of ISG15.Methods:1.transfecting plasmid expressing shRNA into Hep3 B cells.2.Measuring the varieties of cell proliferation before and after transfection by CCK-8 assay;testing the varieties of cell migration and invasion before and after transfection by Transwell cell and artificial basement membrane;testing the changes of cell apoptosis and cell cycle before and after transfection by flow cytometry.Results:1.the proliferation ability of Hep-3B cells was significantly inhibited by ISG15interference(P<0.01).2 the expression of ISG15 protein interfered with the migration and invasion of Hep-3B cells(P<0.01).3 the interference of ISG15 protein expression promotes apoptosis(P<0.01),which is mainly early apoptosis.4 after the interference of ISG15 protein expression,Hep-3B cells in S phase increased significantly and G1 phase decreased(P<0.01);combined with the results of proliferation experiment,showed that the cells had blockage of S phase.Conclusion:ISG15 protein is closely related to the occurrence and development of hepatocelluar carcinoma,which can promoto the proliferation,migration,invasion,and depress apoptosis of Hep3 B cells.The abate of expression of ISG15 protein can make a cell cycle arrest in S phase in Hep3 B cells.
