Objective: Established and compared of eleven different source Tibetan medicine "Dida" plants by high performance liquid chromatography fingerprinting.Four different source "Dida" plants as Swertia mileensis,Saxifraga tangutica,Swertia chirayita and Swertia mussotii to anti-liver injury in vivo and in vitro pharmacodynamics,in order to provide a scientific basis for clinical use.Methed: Established eleven different source "Dida" plants fingerprints,Four different source "Dida" plants study on acute liver injury in mice.With CCl4 to establish a model of chemical hepatic injury,Swertia mileensis,Saxifraga tangutica,Swertia chirayita and Swertia mussotii two doses(3 g/kg,6 g/kg),intragastric administration,1 times per day for 7 consecutive days,testing alanine aminotransferase(ALT),aspartate aminotransferase(AST)and total protein(TP),calculation of the indices of liver and liver pathologytissuesections.Four different source "Dida" plants study on chronic liver injury in mice.Mice subcutaneous injection 20% CCl4(2 times a week),to Swertia mileensis,Saxifraga tangutica,Swertia chirayita and Swertia mussotii two doses(1.5 g/kg,3 g/kg),intragastric administration.1 time/day,for 45 days,testing alanine aminotransferase(ALT),aspartate aminotransferase(AST),total protein(TP)and total bilirubin(TBI),the calculated indices of liver and spleen index and index of the pancreas and liver pathology tissue sections.Four different source "Dida" plants in vitro studies on liver damage determined in cell culture supernatants of alanine aminotransferase(ALT),aspartate aminotransferase(AST),malondialdehyde(MDA)content and superoxide dismutase(SOD)activity.Result: Completed eleven different source "Dida" plants building and similarity evaluation of fingerprint patterns.Four different source "Dida" plants study on acute liver injury in mice.4 drugs can significantly reduce the ALT and AST levels(P<0.01),6 g/kg group in Swertia mussotii works best.Four different source "Dida" plants study on chronic liver injury in mice.Four drugs can significantly reduce the ALT and AST levels(P<0.01),improvement of pathological injury of hepatic tissue.Swertia mussotii,Swertia chirayitaeffect than Swertia mileensis.Four different source "Dida" plants in vitro studies on liver damage.Treatment group compared with the control group ALT,AST and MDA content decreased SOD activity promotion.Four species of plants in different concentration on the activity of the different effects of different,there are some differences,ummary display: Swertia mussotii,Swertia chirayitaeffect than Swertia mileensis.Conclusion: This project established a complete set of Tibetan medicine "Dida" high performance liquid chromatography fingerprinting of this method can be used as a basis for its quality control and evaluation.Swertia mileensis,Saxifraga tangutica,Swertia mussotii and Swertia chirayitaon are all have effect at protective the liver.There are some differences in the effects of different concentrations on liver injury in acute and chronic CCl4 injury mice.The effects of different concentrations on L-O2 in human hepatocytes were different,and their mechanism might be related to antioxidation,promote its wider application in clinical.
