ObjectiveExploring which can accurately predict the prognosis of patients with gastric carcinoma(GC) becomes a hot research in our country where the mortality of GC ranks second in various malignancies. The purpose of this work was to detect the expression Lysosomal-associated protein transmembrane 4 beta-35 (LAPTM4B-35) in GC and precancerous lesions and analyze the correlation between expression of the protein and the severity of GC and precancerous lesions, at the same time, to analyze the relationship between expression of the protein and the clinicopathological features and prognosis of patients with GC. Preliminary discussion the role of abnormal expression of the protein in the development of GC and the value in judging the prognosis of GC,to screen reasonable and effective indicators for clinical of early detection of GC and provide a theoretical basis for determining the prognosis in patients with GC.MethodsA retrospective analysis of 157 cases of GC and its follow-up data was implemented. Immunohistochemical staining and Western blot analysis were used to detect the expression of LAPTM4B-35 protein in precancerous gastric lesions and GC.The correlation of abnormal expression of LAPTM4B-35 protein and clinicopathological features of GC patients was analyzed by chi-square test. The gene expression on survival was used by log-rank test and to do univariate survival analysis by Kaplan-Meier survival curves. And establishing multivariate Cox regression mode to analysis independent prognostic factors for survival of GC. P?0.05 was considered statistically significant.ResultsLAPTM4B-35 expression was increased steadily in sequential stages of precancerous gastric lesions. Positive LAPTM4B-35 expression was more frequently detected in patients with distant metastasis (P=0.023) and ?+? TNM stages (P=0.042)in the discovery cohort. Kaplan-Meier survival curves and univariate analysis showedthat expression of LAPTM4B-35 had a significant impact on overall survival of patients with gastric carcinoma in discovery cohort (P<0.001). LAPTM4B-35 expression was an independent prognostic indicator for the overall survival of patients with gastric carcinoma.ConclusionAt present our findings show that LAPTM4B is an important gene in the development of human gastric epithelial cell adenocarcinoma, and the overexpressed LAPTM4B-35 protein is an independent factor which can determine the overall survival for prognosis of GC patients. The activation of LAPTM4B gene and overexpression of LAPTM4B-35 protein may be the key to the process of precancerous lesions, and its detection and intervention may be an important intervention targets to slow progression of precancerous lesions, but also it has important clinical significance to accurately determine the prognosis of patients with GC.
