OBJECTIVE: The phosphorylation of phosphoinositide 3-kinase(PI3K)and protein kinase B(Akt or PKB)downstream of the signal transduction pathway play an important role in the development and progression of human tumor,And has been confirmed that the occurrence of cervical cancer and human papilloma virus(Human Papilloma Virus,HPV)related to persistent infection.The aim of this study was to investigate the expression of PI3 K and AKT in cervical lesions and their correlation with HPV infection.Methods: 1.Collected 17 cases of cervical biopsy and surgical resection of the First Affiliated Hospital of Bengbu Medical College.There were 30 cases of normal cervical tissues,76 cases of cervical intraepithelial neoplasia(18 cases of LSIL,58 cases of HSIL),68 cases of cervical cancer(36 cases of stage I,32 cases of stage II).2.The expression of PI3 K p110? and p-AKT protein was detected by immunohistochemical SP method.3.HPV-DNA was detected by hybridization capture(HC2)in cervical mucus exfoliated cells.4.The data were analyzed by SPSS19.0.The results were analyzed by clinical data.The results were analyzed by Mann-Whitney U test and Spearman rank correlation analysis.P <0.05 was defined as statistically significant.Results: PI3 K p110? protein expression in cervical cancer group was significantly higher than that in LSIL group and HSIL group(P <0.05).The expression of PI3 K p110? protein in HSIL group was significantly higher than that in LSIL group(P<0.01).The expression of p-Akt protein in LSIL group was significantly lower than that in HSIL group and cervical cancer group(P <0.01),but there was no significant difference between HSIL group and cervical cancer group(P> 0.05)There was a positive correlation between p110? protein expression and p-Akt protein expression in cervical lesions(P <0.001).The expression of PI3 K p110? and p-Akt was positively correlated with HPV infection(P <0.05),but the expression of PI3 K p110? and p-Akt was correlated with FIGO staging,pathological grading and lymph node metastasis(P> 0.05).Conclusion: The expression of PI3 K p110?,p-Akt and HPV infection are closely related to the process of cervical lesion.The combined detection of PI3 K p110? and p-Akt may provide the important information for the early diagnosis of cervical lesions.The application of targeted drugs to cervical cancer may provide therapeutic value for cervical cancer.
