[Objective]DDX39 is a member of the DEAD-Box RNA helicase family,which plays an important role in the regulation of gene expression and RNA metabolism in vivo by recognizing innate immune function, regulating intracellular signaling pathways, RNA transcription and cleavage, and regulating nuclear epigenetics.However in the process of regulation of different tumors DDX39 can inhibite or promote tumors. The function of DDX39 in renal clear cell carcinoma is not clear and its correlation with clinical prognosis still needs further study. Through our study, we will find a novel theory for renal cell carcinoma in the formation of proliferation and metastasis .We also have hope to find a new target for renal cancer treatment. And we can explore a new molecular marker for predicting the prognosis outcome of renal cell carcinoma.[Methods]1?We Use the Oncomine biological database to analyze the expression of DDX39 in clear cell carcinoma and its relationship with prognosis.We also use RT-PCR and Western-blot to detect the different expression of DDX3 9 between tumor tissues and adjacent tissues from CCRCC patients, also the different expression of DDX39 between RCC cell lines and normal renal tubular epithelial cells.2?The expression of DDX39 in renal clear cell carcinoma was analyzed by immunohistochemistry. We will analyze the relationship between the expression of DDX39 in renal clear cell carcinoma and the prognosis of patients.3?siRNA (small interfering RNA) knocks down DDX39 form RCC cell lines and plasmid transient transfection overexpresses DDX39 to RCC cell lines. The knockdown and transfection efficiency were verified by RT-PCR and Western-blot. CCK8 method and plate clone experiment will be used to verify the proliferation of DDX39 with low expression and overexpression. Transwell cell migration assay and cell invasion experiment will be used to verify the metastatic ability of DDX39 with low expression and overexpression.[Results]1. The results showes that DDX39 is highly expressed in renal clear cell carcinoma.The expression of DDX39 is significantly different between adjacent tissues and tumor tissues, also significantly different between RCC cell lines and normal renal tubular epithelial cells.2. The expression of DDX39 is correlated with the tumor size, the TNM staging and distant metastasis. The expression of DDX39 is associated with the overall survival and the disease-free survival in patients with renal cell carcinoma by Univariate and multivariate analysis. DDX39 could be used as an independent predicted factor of renal cell carcinoma prognosis.The high expression of DDX39 means the poor prognosis.3. The siRNA and plasmid transient transfection technology can effectively knock down and overexpress the DDX39. In vitro cell function test results show that knocking down DDX39 from the RCC cells, the proliferation and metastasis ability is inhibited, and overexpressing DDX3 9 to the RCC cells,the RCC cells proliferation and metastasis ability is enhanced.[Conclusion]DDX39 in renal clear cell carcinoma shows the characteristics of proto-oncogene. It can promote proliferation and metastasis of CCRCC. DDX3 9 is expected to be a new molecular marker for predicting the prognosis of patients and a novel target for genetic therapy.
