ObjectiveTo investigate the immunomodulatory activity of Ginkgo biloba extract(EGB),spleen lymphocytes and peritoneal macrophage from the mice were taken as target cells in vitro.The immune function model was established for studying the immunomodulatory activity of EGB.In vivo immunomodulatory activity of EGB was detected by the establishment of immunocompromised mice model induced by cyclophosphamide(CTX).These would provide experimental evidence for the development and utilization of EGB.MethodsThe effect of spleen lymphocyte proliferation was detected by MTT method in the different concentration EGB groups.The effect of cytokines secreted was detected by ELISA kits in the different concentration EGB groups.immunosuppressive mice model was established by CTX.The effect of immune organ index was measured by weighing the spleen and thymus in the different concentration EGB groups and immunosuppressive mice model group.The effect of the IL-4,IL-12,IL-13 and IFN-? m RNA gene expression was detected by RT-PCR in the different concentration EGB groups and splenic lymphocytes of immunosuppressive mice model group.ResultsStimulation index(SI)of spleen lymphocyte was improved in the different concentration EGB groups,EGB combined with concanavalin A(Con A)group and or EGB combined with lipopolysaccharide(LPS)group.The SI of splenic lymphocytes in the different concentration EGB groups were higher than EGB + Con A group and EGB + LPS group.The secretion of IL-4,IL-12,IL-13 and IFN-? cytokines in spleen lymphocytes were enhanced in the different concentration EGB groups.After splenic lymphocytes were infected by staphylococcus aureus,EGB can the SI of spleen lymphocytes were enhanced in the different concentration EGB groups.The effects of macrophage devouring staphylococcus aureus were enhanced in the different concentration EGB groups,EGB + Con A group and EGB + LPS group.And The effects of EGB groups were higher than others groups.The spleen index,thymus index and the expression levels of IL-4,IL-12,IL-13 and IFN-? m RNA were increased by using EGB in the immunosuppressive mice model.ConclusionEGB could enhance the proliferation function of spleen lymphocytes.EGB could increase the proliferation of spleen lymphocytes infected with Staphylococcus aureus.EGB could enhance the ability of macrophage devouring staphylococcus aureusEGB could enhance the spleen index and thymus index in immunosuppression mice model,which indicated that EGB had the effect of protecting the immune organs.EGB could increase the expression of IL-4,IL-12,IL-13 and IFN-? m RNA in splenic lymphocytes,indicating that EGB has immunoregulatory effect.
