Effect Of Pterostilbene On Apoptosis Of Tongue Squamous Cell Carcinoma Line Tca8113

Objective To investigate the effect of pterostilbene on Apoptosis of Tongue squamous cell carcinoma line Tca8113 and its related mechanism.Methods Determined by MTT method to detect pterostilbene different concentrations and different action time of tongue cancer cell strains Tca8113 influence of active growth,Use 0,2.5,5,10,15 and 20 mu of pterostilbene mol/L for final concentration of Tca8113 medium tongue cancer cells,using 96-well plates for planking,role respectively after 24 h,48h and 72 h computer detection OD value.After 48 h inverted microscope and flow cytometry to detect Tca8113 cells apoptosis after processing by pterostilbene.The expression of Bax,Fas and Caspase3 protein was detected by Western blot.Using SPSS19.0 software analysis,measurement data to(?)said,between groups using the variance analysis.P <0.05 for the difference was statistically significant.Results Determined by MTT test results showed that different concentrations of pterostilbene Tca8113 of tongue cancer cells growth inhibition,and with the increase of drug concentration pterostilbene,promote apoptosis effect increased.Morphological observation found that pterostilbene action after the cell apoptosis remarkable state,part of the cell cytoplasm enrichment characteristics.Flow cytometry test demonstrate that the effect of pterostilbene Tca8113 in tongue cancer cells,different concentrations of pterostilbene 48 h concentration medium,induction of cell lines total apoptosis rate of the mean difference was statistically significant compared with negative control group(P < 0.05),and with the increase of drug concentration pterostilbene,promoting apoptosis effect gradually strengthen,2.5?mol/L pterostilbene 48 h concentration medium,induction of cell line averages 26.76% of total rate of apoptosis,20?mol/L pterostilbene trained 48 h,inducing cell line averages 91.11% of total rate of apoptosis.Determination of Western blot showed,Bax,Fas,enhanced Caspase3 protein expression level increased with increasing concentration of pterostilbene.Conclusions Purple stalks can inhibit the growth of tongue cancer cell line Tca8113,upregulate the expression of Bax,Fas and Caspase3 protein,which may be one of its mechanism.
