OBJECTIVE To investigate the expression of NDY1(histone demethylase-1)、EZH2(zeste gene enhancer homologous gene 2)and P16ink4 ain human ovarian carcinoma and the incidence of cancer,the impact of invasion and metastasis,and the relationship among the three.In order to provide a theoretical basis for elucidating the potential value of NDY1,EZH2 and P16ink4 a in the clinical diagnosis of ovarian cancer.METHOD1.Specimen collection Ninety five cases of normal ovarian tissue and ovarian tumor of different grades diagnosed were collected from the First Affiliated Hospital of Guangxi Medical University’s patients from 2010 to 2014.The HE-stained slides of these cases were reviewed and a concordant agreement was established for the confirmation of the diagnosis between three trained pathologists,based on FIGO staging 2014.2.Immunohistochemical method The expression of NDY1,EZH2 and P16ink4 a protein in 20 cases of normal ovarian tissue,25 cases of benign ovarian tumor and 50 cases of ovarian malignant tumor were detected by immunohistochemistry,the titers of NDY1,EZH2 and P16ink4 a monoclonal antibodies were explored using sterile PBS buffer as a control.3.Microscopic observation The expression of NDY1,EZH2 and P16ink4 a in normal ovarian tissue,benign ovarian tissue and ovarian malignant tumor tissues were observed under200-fold and 400-fold microscopy,respectively.4.Statistical analysis Correlation analysis was used to determine the correlation among NDY1,EZH2 and p16 protein expression,and the difference was statistically significant according to the level of α= 0.05 and P<0.05.RESULT1.The positive expression rates of EZH2 protein in ovarian cancer tissues,benign ovarian tumor tissues and normal ovarian tissues were 76%,48% and20% respectively,and was significantly higher in ovarian cancer tissues(P<0.05).Overexpression of the EZH2 was correlated with tumor grade,FIGO stage and lymphatic metastasis(P<0.05),but there were no correlation with patients’ age(P>0.05).2.The positive expression rates of NDY1 protein in ovarian cancer tissues,benign ovarian tumor tissues and normal ovarian tissues were 48%,40% and25% respectively,and was significantly higher in ovarian cancer tissues(P<0.05).Overexpression of the NDY1 was correlated with tumor grade and FIGO stage(P<0.05),but there were no correlation with patients’ age and lymphatic metastasis(P>0.05).3.Expression of P16ink4 a was more frequent in normal ovarian tissue compared with benign tumor and ovarian cancer(P<0.05),the positive expression rates were 48%,72% and 95% respectively.P16ink4 a down-regulation was correlated with tumor grade and FIGO stage(P<0.05),but there were no correlation with patients’ age and lymphatic metastasis(P>0.05).4.Moreover,EZH2 positive rate was negatively correlated with expression of p16 protein in ovarian cancer(r=-0.326,P<0.01).EZH2 positive rate was positively correlated with expression of NDY1 protein in ovarian cancer(r=0.736,P<0.05).CONCLUSION1.The positive expression rate of EZH2 and NDY11 in ovarian cancer tissues was significantly higher than that in normal ovarian tissues and benign ovarian tumors,in contrast,the expression of P16ink4 a in ovarian cancer tissues was significantly lower than in benign ovarian tissues and normal ovarian tissues.Indicating that NDY1,EZH2 and P16ink4 a protein may be involved in the development of ovarian cancer.2.Three of them were significantly associated with tumor grade,FIGO stage and lymphatic metastasis,indicating that NDY1,EZH2 and P16ink4 a may involve in the progression and metastasis of ovarian cancer.3.Combined detection of NDY1,EZH2 and P16ink4 a protein expression in ovarian malignant tumor tissue may help to understand the malignant degree of ovarian cancer in more detail.While the three may also serve as an effective target for new tumor gene therapy.
