Objectives: This topic is to compare the main distribution and the similarities and differences of nasopharyngeal bacterial community structure between the healthy people and patients with nasopharyngeal carcinoma(NPC),and also to reveals the influence of radiotherapy for nasopharyngeal carcinoma patients of nasopharyngeal symbiotic bacteria;As well as to explore dynamic changes and rules of nasopharyngeal symbiotic structure sequence in radiotherapy.Method: 9 healthy volunteers?7 patients with NPC aged 22 ~ 56,KPS score > 90,The object of study with the users who don't have the history of antibiotic,hormone and immune inhibitor in recent one month;The NPC patients treated with intensity modulated conformal radiotherapy technique,dose of radiotherapy: 6840-7400cGy/30-33 f,treated about 6weeks.Healthy people were taken by 9 nasopharyngeal swab samples;14nasopharyngeal swab samples were collected for every patient.After the DNA samples were extracted by using T-RFLP technology to analysis,we can get the datas of nasopharyngeal bacterial community structure of T-RFLP from the healthy people and the patients with NPC in the process of and nasopharyngeal carcinoma radiotherapy.Results:1.There are obviously differents in nasopharyngeal bacterial flora distribution structure between the healthy people and patients with NPC.The number of NPC patients' T-RF is significantly higher than the healthy people and the number is statistical(P<0.05).1)Healthy people can get 82 different terminal restriction fragments in a range from 56 bp to 493 bp.The top 10 advantage T-RFs are T-RF119?TRF399?T-RF235?T-RF 400?T-RF 402?T-RF152?T-RF109?TRF393?T-RF 57?TR-F 488.2)The patients with NPC can get 91 different terminal restriction fragments in a range from 57 bp to 496 bp,and the top 10 are T-RF400?T-RF 62?T-RF77?T-RF 333?T-RF146?T-RF 366?T-RF 235?T-RF 64?T-RF 4013)At the same time we found that nine dominant T-RFs were not only exist in healthy people but also in the patients with NPC: those terminal restriction fragments for62 bp?64 bp?65 bp and 400 bp in patients with NPC have a rising trend,and the 62 bp(333%),400(116%)are the most obvious;In contrast,the dominant T-RFs for 399 bp(86%),402(75%)are the largest decline.4)According to the analysis of diversity indexes,We found that the occurrence of NPC can enhance the population of nasopharyngeal bacterial,which could make the bacterial species increased obviously(10.9%)and also make the diversity of flora structure increased significantly(7.9%),but the Species distribution evenness variation is declining(7.3%).2.The nasopharyngeal flora composition and distribution of NPC patients are in a state of dynamic changes in the process of radiotherapy:1)The distribution trend of 22 kinds of predominant groups of bacteria in of NPC patients were: The 8 kinds of T-RFs was falling and 2 kinds of T-RFs was growing and the rest of T-RFs represented by the bacterium were not a long stay in nasopharyngeal region.2)In Bacteria diversity indexes,Shannon-Weiner Index?Simpson index,the number of T-RFs before radiotherapy are the highest.The scope of bacterial group similarity coefficientin different radiotherapy periods are between 0.015 ~ 0.015.The nasopharyngeal flora similarity in the second week and at the end of the treatment is the highest whose number is 0.99.But the nasopharyngeal flora similarity before treatment and the 4th week is the lowest similarity,which is 0.015.Combining with clustering analysis between samples of different periods,We found that a random dynamic change of Nasopharyngeal carcinoma flora structure.To the end radiation therapy,the number of the T-RFs decline for 46%,Shannon index fell by 5%,Evenness index increased by 5%,but the Simpson index change is not obviously.Conclusion:1.Compared with the healthy people,the NPC patients' nasopharyngeal flora structure and the distribution of advantage bacterium group are obviously different.Through comparing with healthy people's diversity index,we found that the number of the NPC patients' nasopharyngeal bacterial groups is more bigger,community structure is more complex and more diverse,species distribution uniformity is more poor.2.In the process of radiotherapy,the nasopharyngeal flora structures in NPC patients have a random change over time.The radiotherapy can restrain the growth of the symbiotic bacterial in nasopharyngeal carcinoma,make the bacterial species decrees significantly,and the complicated and diversified degree of the NPC patients' bacterial community structure is declining.Maybe the changes of the NPC patients' advantage fragment distribution have certain reference significance on radiotherapy evaluation and relate infectious complications development process.
