| Objective: 1?To observe the acute toxicity of Jianpi Anchang Pill on mice twice a day(interval 6h);2?To establish the model that stagnation of liver qi and spleen deficiency in rats,and detect the damage to liver and kidney and the content of VIP and SP in plasma;3?Collect colon tissue,and detect the contents of VIP and SP,and analyze the protein expression in colon tissue;make the pathological sections of colon tissue and observe it's athological changes.Through the above experiments to analyze the toxicity of Jianpi Anchang Pill and the effect on the level and protein expression of VIP and SP in brain-gut peptide.Methods: 1?The maximum dose of Jianpi Anchang Pill was given to the mice twice a day,and the shape,color,respiration,movement,nose,eye,oral secretions and animal body weight and animal dead time were observed and recorded.At last,the autopsy and pathology of the dead animals were performed;2?To establish the model that stagnation of liver qi and spleen deficiency in rats,and to examine the liver function and renal function and plasma VIP and SP;3?Detect the level of VIP and SP in colon tissue by immunohistochemistry,and observe the expression of VIP and SP protein in colon by HE staining.4?Make the pathological sections of colon tissue and observe it's athological changes.Results: 1?The vital signs of the mices were normal during the two weeks of dosing,and there was no obvious toxicity and death have been found.Compared with the control group,both the weight and food intake was no significant difference(P>0.05).Killed all the mices after the experiment,and checked the animal organs which were all normal,and there was no obvious organic disease been found.Also,there was no significant difference compared with the control group.2?The test results of ALT?AST?Cr and BUN : Compared with the normal controls,there was no statistical difference through the detection of creatinine and urea nitrogen values(P>0.05).The test results of plasma VIP :Compared with the normal controls,model group increased significantly(P<0.05);Compared with the model group,the large,medium and small groups were significantly decreased(P<0.01).The test results of plasma SP : Compared with the normal controls,model group increased significantly(P<0.05);Compared with the model group,the large,medium and small groups were significantly decreased(P<0.01)3?(1)The results of VIP in the colon tissue : Compared with the normal group,the expression of VIP was significantly rised in model group and medium? small dose group(P<0.05);Compared with the model group,the large group and the positive groups were significantly rised(P<0.05),the medium and small dose groups were significantly decreased(P<0.05).(2)The results of SP in the colon tissue :Compared with the normal group,every groups were rised,the small group and the positive groups were rised remarkable(P<0.05),however,large and medium groups rised much more remarkable(P<0.01).Conclusion:Jianpi Anchang Pill have been proved to be low toxicity and safe,and it can significantly reduce brain-gut peptide VIP and SP levels in plasma,and inhibit the expression of VIP and SP protein in colon tissue,and relieve the inflammatory degree.So as to promote the recovery and improvement of the symptoms of D-IBS. |
