ObjectiveThrough the optimization of fructus schisandrae ultrasonic extraction conditio ns,researching of quality standard,researching of fructus schisandrae fingerprint,determining the optimal northeast three provinces different harvest time of origin and the best harvest time;Through the study of fructus schisandrae pharmacodynamics,based on histopathological observation for the treatment of gastric ulcer in rats,providing the basis for study of fructus schisandrae in the treatment of gastric ulcer of experimental support.MethodsUltrasonic extraction was used to fructus schisandrae alcohol armour and the percentage of fructus schisandrae in measurement indicators,using the multi-factor orthogonal experiment to the investigation;Of fructus schisandrae alcohol armour as reference substance,using high performance liquid chromatography(HPLC)method for determining the content of fat tasty ZiMu element;Using the HPLC method established the three northeastern provinces of different harvest time of schisandra fruit fingerprint chromatography conditions;Using tissue pathological observation and weight of rats to determine of fructus schisandrae extracts in the treatment of rats.ResultsUltrasound extraction conditions of optimization:the primary and secondary order of the influence of various factors:ethanol concentration(%)>solid-liquid ratio>ultrasonic time(min)>extraction times,ultimately determine the best extraction condition for:(1:1 0),the ratio of material liquid ultrasonic extraction with 75%ethanol solution,time for 30 min;Quality criteria:different harvest time the content of fructus schisandrae alcohol armour each are not identical,but the overall trends,namely:mature fruiting>half mature fruiting stage>total;Different regions have different content of fructus schisandrae alcohol armour,namely:jilin city>liaoning jinzhou qinghe>heilongj iang yichun heilongjiang province;Fingerprint study:chromato graphic conditions:Spherisorb-C18 chromatographic column(4.6mm×250mm,5 μm),mobile phase:A water,B methanol.Gradient elution:0～5min,50～60%B;5～25min,60～70%B;2 5～45min,70～80%B;45～5 5min,8 0～80%B;55～60min,80～1 00%B;60～70min,100～100%B;Flow rate:1.0 ml/min;detection wavelength of 250nm;Column temperature 25 ℃;Sample quantity:10 mL;Test time:7 0 m i n.A t the same time,examined the fingerprint of stability,precision and repeatability,the results show that the relative retention time of common peak RSD(%)is less than 3%,conform to the rules.The experiment of fructus schisandrae alcohol armour for the controlled peak,determines the relative retention time of 13 common peak is stable,but the peak area is different;Pharmacodynamics study:extracts from fructus schisandrae can better treatment method of acetic acid gastric ulcer in rats.ConclusionThe results showed that fructus schisandrae alcohol armour content has been increased over harvest time,namely:mature frui.ting the highest content of fructus schisandrae alcohol armour,for later provide a basis for the study of different harvest time of fructus schisandrae chinensis.In addition,the three northeast provinces fructus schisandrae,heilongj iang,jilin,liaoning,jilin has the highest content of origin,jilin dunhua is suitable for planting in large quantities of fructus schisandrae,Extracts from fructus schisandrae of pharmacodynamics experiment results show that the fruit of Chinese magnoliavine has good treatment effect to gastric ulcer in rats,as the fruit of Chinese magnoliavine in the treatment of gastric ulcer research some experimental basis will be provided.
