Objective:The aim of this study is to investigate the effect of BMP-2 on the proliferation and cell cycle of MG-63 cells with the affection of the ERK signal pathway and to analyse whether the BMP-2 involved in the biological effects on MG-63 cells affected by the ERK signal pathway and explore the biological effects of interaction between the ERK signal pathway and the BMP-2 in vitro.Methods:The blocking agent of ERK signal pathway(PD98059)was added to block the ERK signal pathway and then the BMP-2 was used to stimulate MG-63 cells.The mitogenic potential of MG-63 cells was measured by MTT;the impact of the cell cycle was measured by flow cytometry.Results:The results of the MTT indicated that proliferation of MG-63 cells affected by the BMP-2 was significant inhibited by the block of the ERK signal pathway;The results of the flow cytometry cycle analysis showed that the BMP-2 involved in cell cycle regulation of cells which affected by the ERK signal pathway.Conclusion:Our experiments verified that the BMP-2 plays an suppression role in the proliferation of the MG-63 cells and it restrains the proliferation of the MG-63 cells which was treated by the ERK signal pathway.The BMP-2 involved in the regulation of the MG-63 cell cycle by The ERK signal pathway. |
