In this paper,Chinese herbal medicine Brucea javanica and Brucea javanica oil grease injection were used as material to study.A high-performance liquid chromatographic method with an evaporative light-scattering detector has been developed for the separation and quantitative analysis of underivatized long chain fatty acid in Brucea javanica and Brucea javanica oil grease injection.An isocratic elution mode using methanol/water/acetic acid(88:11:1,v/v/v)as mobile phase with a flow rate of 1.0 mLˇmin-1 was used.The separation was effected by using an Agilent Eclipse XDB-C18 analytical column(4.6x150 5 ?m).ELSD drift tube temperature was set at 70?.Calibration curves of the oleic acid was well correlated within the range of 0.5-10 mg-mL-1.HPLC-ELSD has been developed for the characteristic fatty acids analytical fingerprints of the raw materials and oil grease injection.The characteristic analytical fingerprints of the Hydrolyzate of Brucea javanica oil and oil grease injection were carried out on the HPLC method for analysis oleic acid above.10 batches of samples were analyzed under the same HPLC conditions.Analysis on an Agilent Eclipse XDB-C18(4.6×250 5 ?m)column with an evaporative light-scattering detector and eluted with mobile phases containing acetonitrile/isopropanol/hexane.(66:20:14,v/v/v)And data of 10 batches of Brucea javanica and oil grease injection samples were analysed by "similarity evaluation for chromatographic Fingerprint of Traditional Chinese Medicine" software.The chemical components of Brucea javanica oil were analysed by HPLC-MS.Nine common peaks were selected in chromatograms,eight of them were identified in chromatograms based on their MS data and all the common peeks were separated effectively.The precision,repeatability,and stability of this method were satisfying.The proportion of fatty acid composition and extraction rate of Brucea oil extracted by different method were studied.The result shows that the method of ultrasound-assisted and enzymatic assisted take no impact on fatty acid composition.Choose the enzymatic assisted method as the best extract method,single-factor extraction tests and response surface analysis were employed to determine the optimum technological conditions.The results showed that optimum extraction conditions were as follows:4.64%enzyme amount,enzymatic treatment was performed 3.48 h under 45.84?.
