Obesity has become a global epidemic,serious harm to human health.Obesity and overweight patients are often accompanied by a variety of complications: insulin resistance,dyslipidemia,hypertension,type 2 diabetes and nonalcoholic fatty liver Incidence and mortality increased.The treatment of obesity includes: regular lifestyle,medication and weight loss surgery.But its traumatic and risk and strict surgical indications,and therefore not suitable for all obese people.Due to a large number of side effects,traditional weight loss drugs applications are limited,such as: Fenting: blurred vision,difficulty breathing,primary pulmonary hypertension;Amphetamine: common side effects are dry mouth and insomnia;Orlycece: fat diarrhea incontinence and oily stool;Sibutramine:There is a risk of a stroke,heart attack,so looking for good results,fewer side effects of drugs is the most important treatment of obesity.Liraglutide is a synthetic analog of glucagon-like peptide-1,which has the effect of lowering blood glucose,improving insulin sensitivity,controlling body weight and improving blood lipids.Therefore,liraglutide is useful for the treatment of obesity and its associated complications has a broad application prospects.The aim of this study was to observe the effects of liraglutide on body weight,body fat,blood glucose and liver in obese mice induced by high fat diet.Thirty-nine male C57 BL / 6J mice were randomly divided into two groups: normal diet group(SF group n = 13)and high fat diet group(HFD group n = 26).at 8 weeks of feeding,all mice were measured for body weight.The normal diet control group and high fat diet control group were injected intraperitoneally with equal volume of normal saline,high fat liraglutide group subcutaneous injection(200ug / kg).In the intervention process,weighed twice a week.After 8 weeks of intervention,ITT and GTT were performed.Followed by each group of three mice were randomly sacrificed to observe the mouse body fat,liver weight and related blood biochemical indexes.Effect of liraglutide on body weight and insulin sensitivity in obese mice.After 8 weeks of treatment with liraglutide,the body weight was significantly lower than that in the HFD group(P <0.05).After 20 weeks of high fat feeding,the body weight(39.11 ± 3.62g)of HFD mice was 1.29 times(P <0.05)in SF group mice(30.27 ± 2.48).After 20 weeks of intervention,the body weight was significantly lower than that in the HFD group(30.22 ± 2.76 g vs 39.11 ± 3.62 g,P <0.05).Eight weeks after the intervention,the IPGTT test showed that the blood glucose levels of the three groups were significantly higher than those before injection and reached the peak at 30 minutes.The absolute values of blood glucose in HFD group were significantly higher than those in SF group at 0,30,60,90 and 120 minutes(P <0.05).The absolute value of blood glucose in liraglutide group was lower than that in HFD group at 60,90 and 120 minutes,but the absolute value was higher than that in HFD group at 30 minutes,and the difference was statistically significant.After 20 weeks of intervention,The blood glucose of the three groups was significantly higher than that before injection,and reached the peak at 30 minutes.The absolute values of blood glucose in HFD group were higher than those in SF group at 0,30,60,90 and 120 minutes,respectively(P <0.05)at 30 and 60 minutes.The blood glucose levels in the liraglutide group were lower than those in the HFD group at 0,30,60,90 and 120 minutes after 30 minutes,60 minutes,90,120 minutes,and the IPITT test significant at 60 and 90 minutes.After 20 weeks of intervention,the blood glucose of the three groups was significantly higher than that before the injection(P <0.05).The blood glucose level of HFD mice was significantly higher than that of SF group at 30,60,90 and 120 minutes,and there was significant difference(P <0.05)at 30 minutes(P <0.05)Of the blood glucose were 30,60,90,and 120 minutes lower than those in the HFD group and had a statistically significant difference at 60 and 90 minutes.Effects of liraglutide on body fat and liver weight in obese mice.Compared with SF group,the white adipose tissue,perineal white adipose tissue and brown fat of HFD group increased to 1.94 times,1.89 times,2.48 times,and1.14 timesof SF control group respectively.Compared with HF group,the white adipose tissue,perineal white adipose tissue,epididymis white adipose tissue and brown adipose tissue of liraglutide group were decreased.Compared with SF group,the liver of HFD group increased to the control group 1.20 times;compared with HFD group,liraglutide group mice were reduced liver.Compared with SF group,the white adipose tissue,perineal white adipose tissue and epididymal white adipose tissue and brown fat of HFD group increased to 4.4 times,3.23 times,3.69 times of SF control group compared with SF group(P <0.05).Compared with HFD group,the white adipose tissue,perineal white adipose tissue and epididymis white adipose tissue and brown adipose tissue were decreased in the subcutaneous white adipose tissue of the liraglutide group,and the epididymis of the liraglutide group White adipose tissue was statistically different from HFD group(P <0.05).liraglutide intervention in obese mice liver HE staining:The lipid in the liver of the HFD group was significantly higher than that in the SF group.The content of lipid droplets in the liver of the liraglutide intervention group was significantly lower than that in the HFD group.4.Effects of liraglutide on blood biochemical parameters in obese mice:Compared with SF group,the levels of alanine aminotransferase(ALT),uric acid(UA),bile acid(TBA),blood glucose(GLU),total cholesterol(TC),triglyceride(TG),High density lipoprotein cholesterol(HDL-c),low density lipoprotein cholesterol(LDL-c)and free fatty acid(FFA)were increased,Compared with HFD group,the level of bile acid(TBA)and blood glucose(GLU)increased and the rest decreased.After 20 weeks of high fat feeding,liraglutide reduced blood glucose levels in obese mice,reduced triglyceride,total cholesterol,low density lipoprotein cholesterol.Liliprootide can reduce the weight of subcutaneous and visceral adipose tissue in obese mice induced by high fat diet.while also reducing the fasting glucose in obese mice induced by high fat diet,improve mouse insulin sensitivity.Liliprootide can reduce the level of triglyceride,total cholesterol,low density lipoprotein,thereby improving the level of lipid metabolism.
