Determination Of Content Of Morusin In Morus Notabiliand Its Inhibitory Effect On ECA109 And Du145 Cell Lines

Morusin is widely distributed in various parts of mulberry,a isoprenyl-based flavonoid,which has antibacterial,antioxidant,analgesic,hypoglycemic,anti-HIV and anti-tumor and other biological activity.A new flavonoids?notabilisins K?and four known flavonoids?morusin,mulberrofuran,neocyclomorusin and mornigrol?were isolated and identified from the branches of Morus notabili by 95% ethanol.The cytotoxicity of each component on five tumor cells?HCT-116,HepG2,BGC-823,NCI-H1650 and A2780?showed that morusin has obvious inhibitory effect on proliferation of five tumor cells.In this paper,the content of total flavonoids in Morusnotabili was determined by spectrophotometry,and the content of morusin was determined by HPLC.In this study,we selected ECA109 cells and Du145 cells as the object of study,take morusin fortest drug,to study the inhibition of these two tumor cells,and to explore its Anti-tumor mechanism.The results showed that the content of morusin in the samples was 1.07 mg·g^-1,the content oftotal flavonoids was 31.69 mg·g^-1,and the content of morusin in total flavonoids was 3.38%.The extraction efficiency ofethyl alcoholwas the highest in different extraction solvents.After administration,ECA109 cells and Du145 cells appeared similar to the phenomenon of "perforation",cell shrinkage circle,adherent cell reduction,chromatin condensation,organelles destruction and the emergence of many cavitation structure,IC₅0 was 16.9?mol·L^-1 and 16.5?mol·L^-1,respectively.Morusin significantly attenuated the expression of Gli1 and VEGF protein in the cells?P <0.05?,which induced ECA109 cells and Du145 cells apoptosis,and with the increase of the concentration of action and the effect of prolonged effect more obvious.It can be concluded that the content of morusin in Morus notabili is more abundant,and ethylalcohol is the most effective extraction solvent.Morusin can inhibit the growth of ECA109 cells and Du145 cells,may inhibit the activation of Hh signaling pathway by reducing the expression of Gli1 and VEGF,thereby inducing tumor cell apoptosis.The results of the study have a certain social value.
