[Objective]Detection of patients with endometriosis (EMs)Serum and ectopic endometrium in endometrial tissue (ovarian endometriosis cyst) optic neuropathy induced autophagy related genes (Optineurin OPTN), P62 protein expression, Comparative OPTN, P62 in patients with endometriosis and the different expression in the normal control group, in order to understand OPTN, P62 relations with endometriosis,explore autophagy genes involved in the role in the pathogenesis of endometriosis for etiological basis, looking for new ideas for treatment ?[Method]1. Using enzyme-linked immunoassay (Elisa) determination of subjects in serum OPTN, P62 protein expression.2. Using the method of real-time fluorescent quantitative PCR (rt-pcr) in patients with EMs of the reign of ectopic endometrium, endometrial tissue and normal endometrial tissue OPTN, P62 mRNA expression.3. The use of Westren Blot method EMs of the reign of patients with endometriosis,endometrial tissue and normal endometrial tissues OPTN, P62 protein expression.[Results]1. The OPTN protein expression in patients with EMs in the serum is higher than normal control group expression level in the serum (P < 0.05),P62 protein in patients with EMs said in the serum is lower than the normal control group expression level in the serum (P < 0.05).2. OPTN mRNA in experimental group of ectopic endometrium tissue, are lower than the control group expressed in endometrial tissue in the endometrium (P < 0.05);P62 mRNA in experimental group of ectopic endometrium tissue, higher than the control group expressed in endometrial tissue in the endometrium (P < 0.05);But the experimental group of ectopic endometrium tissue and in endometrial tissues OPTN mRNA and P62 mRNA expression level had no statistical significance (P > 0.05).3 . The OPTN protein in the experimental group of ectopic endometrium tissue, in the expression of endometrial tissue were lower than control group in the endometrium (P< 0.05);P62 protein in the experimental group of ectopic endometrium tissue, in the expression of endometrial tissue of endometrium is higher than the control group (P <0.05), but the experimental group of ectopic endometrium tissue and protein in endometrial tissues OPTN and P62 protein expression level had no statistical significance (P > 0.05).[Conclusion]1. The OPTN positively related to the autophagy, P62 is negatively related to the autophagy, consistent with the literature.2. The OPTN high protein expression in serum in the EMs group, low P62 protein expression, explain EMs autophagy activity in serum is autophagy activity of normal control group.3. In EMs group organization OPTN, low OPTN mRNA expression and protein P62 protein,P62 high mRNA expression,ectopic patients with EMs,in ectopic tissues autophagy activity than normal endometrial tissue autophagy activity is weak.4. Autophagy activity differences in each group: in patients with normal endometrium tissues,EMs,ectopic endometrium tissue autophagy activity gradually decline.5. This research further confirmed: autophagy is involved in the occurrence and development of EMs, autophagy activity weakened in promoting the local lesions occur, survive, autophagy activity in serum may be the body's immune system and inflammatory reaction in the disease oc-curred in the development of a comprehensive protection mechanism of the body itself.
