Objective: To investigate the expression of Semaphorin4D (Sema4D) in colorectal carcinoma and its correlation with microvessel density (MVD).Methods: We retrospectively analyzed the clinical and pathological data of 61 patients with colorectal cancer undergoing colorectal surgery from 2014 to 2015 in Yunnan Provincial Tumor Hospital. Males:38, Females:23, Median age:55 years old;The expression of Sema4D protein in 61 cases of colorectal cancer tissues and 56 cases of normal intestinal mucosa (5cm or more from the edge of cancer tissue) was detected by immunohistochemical SP method. CD34-labeled microvessels were detected by immunohistochemical SP and microvessel density were counted.Results: Sema4D protein is mainly expressed in the cell membrane and cytoplasm of colorectal cancer tissues, and is negative in the cytoplasm of normal intestinal mucosa.The positive expression rate of Sema4D in cancer tissues [100% (61/61)] was significantly higher than that in normal intestinal mucosa [0 (0/56)],the difference was statistically significant (? 2=113.027, P<0.001) . The correlation between Sema4D and MVD was found in colorectal cancer tissues, and the Spearman correlation coefficient r = 0.333 and p = 0.009, which showed significant positive correlation. MVD expression in the Sema4D expression between the high and low groups for t test analysis,p=0.003 (p<0.01),the difference was statistically significant.Conclusion: Sema4D is highly expressed in colorectal cancer and has a positive correlation with MVD. MVD increases with the expression of Sema4D, suggesting that Sema4D is expected to be a potential target for colorectal cancer.
