Objectives:In this study,the in-situ composite and subsequent compound were to use in manufacturing AgNPs antibiotics on the PCL/SF electrospinning fiber membrane,to expand the application of the membrane.The effectivness of ultraviolet light,on the generation of AgNPs antibacterial agent was also investigated.Both of the polymers of polycaprolactone and silk fibroin,could be used as stabilizers,preventing the aggregation of the AgNPs.Methods:Firstly,solution of PCL/SF = 7/3 proportion,was dissolved in hexafluoroisopropanol,and then silver nitrate were added respectively,at Owt%,0.1 wt%,0.5wt%,1.0wt%,2.0wt%,dissolved 9 h;and then the fiber membrane was fabricated under high voltage electrostatic spinning.The' 1.0wt%AgNO3PCL/SF electrospinning film was divided into 3 groups at the same size,with an uv exposure of Omin,30 min,60 min,120 min.The ratio of PCL/SF = 7/3,PCL/SF = 5/5,PCL/SF = 3/7 were electrospinning,and the optimum electrospinning ratio was selected.The ratio of PCL/SF = 7/3 was electrospined,and the prepared electrospinning films were divided into three films with equal thickness.They were soaked in 1.0wt%aqueous solution of silver nitrate for 3 h,and then taken out with uv exposure of 30min,60min,120min.By SEM,EDX,FTIR,XRF,XRD,the properties of the electrospinning films,were observed,whether there were formation of silver nanoparticles and AgNPs on the membrane.Datas used SPSS23.0 statistical software for statistical analysis.Results:In situ composite method of the preparation of AgNPs concentration was less,which proved the limit of silk fibroin reduction ability.The auxiliary ultraviolet light could improve the concentration of AgNPs.With the the concentration of silver nitrate from 0wt%-2.0wt%,Fiber diameter difference was statistically significant(P<0.5).Fiber diameter from 0.872±0.246 um reduced to 0.647±0.274um.Ultraviolet light from different time nano silver particle size difference was statistically significant(P<0.5).Nanosize silver particles from 45±13 nm diameter increased to 97±29nm.With the longer the illumination time,nanoparticles diameter were higher.(2)When the concentration of silver nitrate was certain,the parameters of electrospinning were adjusted to 12-15cm distance,22kv voltage,2.0ml/h pump rate,10%solution concentration.It was found when the ration was PCL/SF=7/3,the best product could be obtained.Because these resulting fiber morphology was better,and the diameter was uniform.There was not obvious droplets,and the spinning process went smoothly.(3)Subsequent composite approach successfully prepared AgNPs/PCL/SF membrane and the preparation of AgNPs concentration was higher.SEM showed that large amounts of silver nanoparticles assembled on the fiber surface.With an uv exposure of 30 min,60 min,120 min,AgNPs size difference was statistically significant(P<0.5).Nanosize silver particles diameter from 56±23 nm increased to 147±69nm.With the longer the illumination time,nanoparticles on the fiber surface were better.Conclusions:This topic by using the method of in situ composite prepared AgNPs/PCL/SF electrospinning membrane successfully.With silver nitrate concentration increasing,the spinning fiber was fine,and the concentration of silver nanoparticles prepared was higher.Ultraviolet light prolonged,the nanosize silver particles diameter increased.Through fixed silver nitrate concentration 1.0wt%,we choosed the optimal ratio of PCL/SF = 7/3.We prepared with subsequent composite method AgNPs/PCL/SF electrospinning film.The longer ultraviolet light,the greater the nanosize silver particles diameter.Finally nanosize silver particles occurred together.Thus by ultraviolet light time and silver nitrate controlled the size of AgNPs.Two methods in situ composite and follow-on composite preparation of AgNPs distribution,particle diameter and concentration had certain difference,which could do reference for subsequent experimental or clinical demand.
