Objective:Set up SD rat MCC(mandibular condylar chondrocytes,MCC)and NSC(nasal septal chondrocyte,NSC)in vitro models through the static magnetic fields,observes the differences and effectson on chondrogenesis of MCC and NSC under the static magnetic fields.providing guidance for further study on the effect of Flrt-2 on cartilage formation under static magnetic field.For more in-depth study of theoretical basis in maxillofacial growth mechanism in the development proceess differences in chondrogenesis of MCC and NSC in the static magnetic fields,the following provides guidance.Methods:The primary MCC and NSC of 1-3 days neonatal SD rats with mechanical separation-enzymatic digestion combined method were cultured and identified in the same culture conditions.They were underwent 280mT static magnetic fields after most cells attachment,Observed the expression differences of GAG(glycosaminoglycan,GAG)under static magnetic fields by Alcian blue staining.Chondrocytes growth curves were drawed by CCK-8(Cell Counting Kit-8,CCK-8)assay,observing the proliferation differences of MCC and NSC under static magnetic fields.Scratch experimental observation of MCC and NSC migration after 24 hours under static magnetic fields.We detect the expression differences of Flrt-2 and N-cadherin protein with WB(western blot,WB)after the static magnetic fields expose.Results:The same digestion and culture conditions successfully cultured the primary MCC and NSC,Type ? collagen immunohistochemical staining and Alcian blue staining were positive,and the cultured cells were the desired chondrocytes.Two chondrocytes were divided into blank group and experimental group,experimental group were underwent 280mT static magnetic fields after most cells attachment.We tested the positive expression and cartilaginous nodule formation on day 1,3,5,8 by Alcian blue staining,The result is that NSC were stronger than MCC at the same time points,and two group with static magnetic fields were stronger than control group in all assay.CCK-8 assay detected the two cartilage growth curves were basically alike,proliferation ability of NSC were stronger than MCC.Two chondrocytes were scratched after 24h,number of NSC migrated to the scratched area were significantly higher than MCC,indicating that migration ability of NSC were stro-nger than MCC.Two chondrocytes' ability of proliferation and migration were stronger after the static magnetic fields expose.Two chondrocytes on day 1,3,5,8,protein expression of Flrt-2 and N-cadherin were detected,found that the protein expression of Flrt-2 decreased,and N-cadherin increased in the agglutination stage.And then Flrt-2 decreased,N-cadherin increased after the agglutination stage.The protein expression of Flrt-2 were increased after the static magnetic fields expose,While,the protein expression of N-cadherin were decreased.Conclusions:1.Positive expression of NSC were stronger than MCC by Alcian blue staining,and two group with static magnetic fields were stronger than control group in all assay.2.NSC migration and proliferation ability were stronger than MCC,and these ability of two chondrocytes were stronger after the static magnetic fields expose.3.The protein expression of Flrt-2 decreased,and N-cadherin increased in the agglutination stage.And then Flrt-2 decreased,N-cadherin increased after the agglutination stage.The protein expression of Flrt-2 were increased after the static magnetic fields expose,While,the protein expression of N-cadherin were decreased.
