[Objective]To dynamically observe the staining of lymph nodes of larynx and neck of rabbits by injecting carbon nanonarticles,methylene blue and Chinese ink into glottis and supraglottic area in laryngeal cavity of living rabbits,to compare cervical lymph node staining and to trace effect of three kinds of lymph node tracers mentioned above that have been administered in different areas of laryngeal cavity so as to provide theoretical and experimental basis for the clinical application of related lymph node tracer in laryngeal carcinoma and its lymph node metastasis.[Methods]Thirty New Zealand white rabbits were randomly divided into 6 groups(n = 5).The rabbits were anesthetized,injected with carbon nanonarticles,methylene blue and Chinese ink into glottis and supraglottic area with laryngeal needle per os respectively under the guidance of electronic laryngoscope.After the laryngoscope was withdrawn,the cervical lymph node dissection was performed in the experimental rabbits,and the damage to the laryngeal lymphatic network was minimized.The staining number,staining time and stain fading time of laryngeal sentinel lymph nodes in the experimental rabbits were observed dynamically and analyzed.[Results]:The total number of stained lymph nodes in carbon nanonarticles group was 36,followed by methylene blue group(33)and Chinese ink group(7).For strainers administered at supraglottic area:The average number of stained lymph nodes was(5.400 ± 1.140)in methylene blue group,(7.400 ± 1.140)in carbon nanonarticles group of,statistically significant difference was found between the two groups(P<0.05);the staining time was(20.400 ± 1.673)min in methylene blue group and(31.200 ± 2.864)min in carbon nanonarticles group,statistically significant difference was found between the two groups(P<0.05).For strainers administered at glottis:the detected number of lymph nodes was(1.200±0.447)in methylene blue group and(1.800 ± 0.837)in carbon nanonarticles group,no statistically significant difference was found between the two groups(P = 0.305);the staining time was(102.200 ± 3.564)min in methylene group and(86.600 ± 2.702)min in the carbon nanonarticles group,and the difference between the two groups was statistically significant(P<0.05).[Conclusion]The laryngeal sentinel lymph nodes in the experimental rabbits were stained in both carbon nanonarticles group and methylene blue group,the numbers of stained lymph nodes in the carbon nanonarticles groups more than methylene blue group,and the staining time was different.Overall,both of these two methods have pluses and minuses.Chinese ink,however,had poorer effect either in the number of stained lymph nodes or staining time when compared with carbon nanonarticles and methylene blue,so it is not a proper tracer for laryngeal lymph node in rabbit.Pathological examination of the specimen is not affected after the tissue was stained by these three kinds of lymph node tracer.
