Objective: Vascular calcification is a common problem among diabetes,end-stage renal disease and heart failure.Recent progresses suggest that vascular calcification is the calcification of tunica media,which is similar to bone formation;however,its definite mechanism remains unknown.Vascular smooth muscle cells(VSMCs)are the basis in the study of arterial calcification.Long non-coding RNA(lncRNA)refers to the non-coding RNA length of more than 200 nucleotides,it does not have the function of transcription and translation,but it can be combined with RNA or DNA specifically,affecting the transcription and translation.Anti-differentiation non-coding RNA(ANCR)is a kind of lncRNA,ANCR can inhibit the differentiation of epidermal cells,and artery calcification is osteoblastic differentiation of vascular smooth muscle cells(VSMCs).At present,there has no study of arterial calcification and ANCR.To investigate the effects of the anti-differentiation non-coding RNA(ANCR)on the osteoblastic differentiation of vascular smooth muscle cells and explore its possible mechanism.Methods: 10 mM of beta-glycerophosphate(?-GP)was used to induce osteoblastic differentiation of mouse VSMCs,which was infected by lentivirus vectors over-expressing ANCR.The m RNA expression of Run related transcription factor 2(Runx2),bone morphology protein-2(BMP-2)and ANCR were detected by quantitative polymerase chain reaction(qPCR).The protein expression of Runx2,BMP-2 and nuclear factor kappa B(NF-?B)p65 subunit were determined by Western Blotting.The formation of mineralized nodules was determined by Alizarin Red Staining.Results: 1,The expression of Runx2 and BMP-2 and the formation of mineralized nodules stained by Alizarin Red were significantly increased in mouse VSMCs stimulated by ?-GP(P<0.05).2,The expression of ANCR was significantly decreased in the mouse VSMCs stimulated by ?-GP(P<0.05).3,Over-expression of ANCR significantly reduced the mRNA and protein expression of Runx2 and BMP-2 in ?-GP-stimulated VSMCs,decreased the protein expression of NF-?B and attenuated the formation of mineralized nodules(P<0.05).Conclusion: ANCR may attenuate the osteoblastic differentiation of VSMCs through inhibiting the NF-?B signaling pathway.
