Microbial secondary metabolic have been proved to be an important source of hypoglycemic compounds.In this thesis,P.thymicola IBT 5891 was chosen to study its secondary metabolites.A series of compounds were obtained and identified by modern chromatography and spectroscopic techniques,and their potential hypoglycemic activity was exploredThe thesis was divided into four parts.In the first chapter,the structure of the secondary metabolites from genus of Penicillium and the research progress of diabetes with fungi were reviewed.The significance and basis of this research were proposed.In the second chapter,as guided by OSMAC strategy and screened by the production which exhibited UV absorption in HPLC,the reasonable medium of 9 was selected.In the third chapter,as guided by the in vitro ?-glucosidase inhibitory assay,seventeen compounds were isolated and identified from the ethanol extract of P.thymicola IBT 5891.Their structures were elucidated by extensive spectroscopic techniques,including two new compounds and fifteen known compounds.In the forth chapter,seventeen compounds of the isolates were evaluated for their in-vitro ?-glucosidase inhibitory activities.As a result,compounds 1-5 and 7-14 exhibited better activity than the positve-effect drug(Acarbose).The study revealed the anti-hypoglycemic effect of Penicillium thymicola from the ?-glucosidase inhibition perspective.The result of this research may offer chemical entities for the development of ?-glucosidase inhibitors.
