Effect Of Fuzheng Huoxue Jiedu Decoction On Tumor Stem Cells-Dependent PMNs On Tumor Metastasis

Background:based on the traditional Chinese medicine "stasis poison" theory,this study based on before the centralizer,qingrejiedu prescription to join blood detoxification medicine,of the centralizer huoxue jiedu,clinical treatment of advanced breast cancer patients have good curative effect,more and more research shows that its effect on promoting tumor recurrence metastasis of the tumor microenvironment,including multiple complex molecular mechanisms and the specific correlation remains to be further research.Of this study is in the early righting detoxification,stem cell research,on the basis of experiment further establish breast cancer cells,breast cancer stem cells and neutrophils co-culture model analysis and research the centralizer huoxue jiedu party transfer the molecular mechanism of inhibiting tumor recurrence.Methods:Cell model is set up,respectively to establish pure breast cancer 4T1,4 T1 and neutrophils(PMNs)group,the breast cancer stem cells(CSLCs)group,CSLCs with PMNs training group,blank control group for pure breast cancer(4T1)group.Observation 4T1 cells,4T1 cells and cultivation in neutrophils,the transfer function of the difference between tumor cells and related molecular mechanism;,respectively,set up the blank group and drug intervention group(gambogic acid group,the curcumin group,fritillaria,first division,implicit tanshinone group)and blank group for the cell model,the other were to join Chinese medicine intervention group.Observe the centralizer metastasis in breast cancer cells of TCM for activating blood detoxification party and molecular mechanism based on stem cells and neutrophils.Detection:4T1 cells,4T1 breast cancer stem cells and neutrophils were extracted.Determined by MTT method is used to detect gambogic acid,curcumin,fritillaria,implicit tanshinone on 4T1 cell proliferation inhibition rate;Transwell methods to detect the transport rate of tumor cells and the centralizer effect of TCM for activating blood detoxification party transfer of breast cancer cells number;ELISA method to detect the cell group,four drugs intervention after the supernatant of related cytokines u-PA?VEGF secretion level;Western blot method to detect the cell group,four drugs intervention after PAI-1,TFPI2 protein expression?Conclusion:The results show that based on "stasis poison" theory of traditional Chinese medicine centralizer huoxue jiedu party on inhibition of tumor cells have a certain value-added killer function,can inhibit tumor cell metastasis,prolong the survival period.It is mechanism may be related to the centralizer huoxue jiedu regulate blood viscosity,reduce the cell factor the u-PA,the expression of VEGF,effect on stem cells and affect the neutrophil differentiation,improve the tumor microenvironment,decrease the rate of cell metastasis.From protein expression results to analyze its mechanism may be related to the centralizer huoxue jiedu party by cutting neutrophils in the tumor microenvironment,the secretion of IL-8 prompted to type conversion,reduce the infiltration of neutrophils,improve the tumor microenvironment,improve blood flow state to reduce the PAI-1 protein expression,improve the TFPI2 protein expression play to promote apoptosis of tumor,the degradation of MMPs,inhibit angiogenesis,inhibiting tumor metastasis.
