BackgroundThe occurrence and development of hepatocellular carcinoma is the result of complex network based on chronic inflammation.It has been found that the accumulation of macrophages in the liver cancer tissue and the chronic non-specific inflammation of the liver which mediated by macrophages can promote the progression of liver cancer.At present,whether downregulation of Macrophage mediated inflammation by anti-inflammatory drugs could prevent the progress of liver cancer remain obscure.However due to the relationship between inflammation and cancer formation in view of the above,the use of anti-inflammatory treatment inhibit the non-specific inflammatory reaction has the possibility to inhibit the progression of liver cancer.ObjectiveWhether anti-inflammatory therapy has the possibility to delay the progression of DEN induced primary liver cancer.Methods1 Choose 56 male Wistar rats were randomly divided into four groups,such as control group,glucurolactone group,NS AIDS(Aspirin)group and steroid group,each group of 14,establish DEN induced and anti-inflammatory drug intervention of liver cancer animal models.2 Some pathological change we will find with the naked eye about the rats liver of each group at the 8th week,10th week,12th week and 16th week,is there a nodule formation,and nodules diameter and number.3 Using HE staining and optical microscopy to observe the rat liver tissue pathological changes of each group at the 8th week,10th week,12th week and 16th week whether it have liver cirrhosis and/or liver cancer formation and the corresponding time.4 Using immunohistochemical to detect the expression of CD34,CD68 and CD206 in rats liver tissue of each group.5 Using western blot to detect the relatively expression of TNF alpha,TGF beta,IL10 and NF-kappaB and HGF in rat liver tissue of each group.6 Using SPSS20.0 statistical software to deal with the above date results,measurement data using mean +/-standard deviation(x±S)expression,multiple sets of comparison between using analysis of variance,variance together when multiple comparisons using LSD method between groups,variance with Dunnett T3 method when not neat,with P<0.05 said the difference was statistically significant.Result1 There have 3 rats died at the 9th,13th and 15th week,so there's only 53 rats did not appear accidental death,all entered the result analysis.2 General macroscopic observation showed that the rats liver tissue of each group began to develop nodules at the 8th week,and nodules number of each group were gradually increase as time goes on,but the nodules number of control group and glucurolactone group in different periods were significantly greater than NSAIDS(Aspirin)group and steroid group,and the number of steroid group was the least.3 HE staining results showed that the rats liver tissue of each group began to develop different degree of liver cirrhosis,liver cancer at the 8th week,but the time of liver cirrhosis and liver cancer of control group and glucurolactone group in early were earlier than NSAIDS(Aspirin)group and steroid group,even the differentiation of liver cancer of control group and glucurolactone group were higher than NSAIDS(Aspirin)group and steroid group,and time of liver cirrhosis and liver cancer of steroid group was the latest,differentiation of liver cancer was the highest.However,at the late time of this experiment showed there were not obvious difference between the groups.4 At the 8th,10th,and 12th week of this experiment the number of positive cells of CD68 and CD206 of rats liver tissue of control group,glucurolactone group and NSAIDS(Aspirin)group were greater than steroid group,but it were not obvious difference between the groups at the 16th week.during the whole experiment,the number of positive cells of CD34 of rats liver tissue of control group,glucurolactone group and NSAIDS(Aspirin)group were greater than steroid group.5 At the 8th,10th,and 12th week of this experiment,western blot test was shown that the relative expression of TNF alpha,IL-10,HGF and TGF-beta of rats liver tissue of control group and glucurolactone group were higher than steroid group.However,the relative expression of such factors of NSAIDS(Aspirin)group was lower than control group and glucurolactone group only at the 8th week.Conclusion1 Chronic inflammation of the liver tissue reaction which was mediated by macrophages can promote the progress of hepatocellular carcinoma.2 Anti-inflammatory therapy can deny the development of hepatocellular carcinoma to some degree which was because it could directly or indirectly inhibit liver inflammation and improve local inflammatory microenvironment at the early time.
