Objectives:A set of ancestry informative SNPs loci by screening and build 27-plex SNP multiple amplification system optimizations based on SNaPshotTM technology.Evaluation of 27-plex SNPs amplification system sensitivity and species specificity,application of 27-plex SNPs multiple amplification system analysis of different populations genetic component and group source,verify the 27-plex SNPs multiple amplification system is concluded that the effectiveness of racial source.Methods:On the 27-plex SNPs amplification system of concentration of PCR primers and extend-primer for further optimization.The system was validated for sensitivity and species specificity.A total of 533 samples from African,Southern China Han,Yi,Hui,Miao,Tibet,Uygur,European,Central Asian,Western Asian,Southern Asian,Southeast Asian and South American were collected.Clustering analysis of 533 genotypes from the above 13 populations was performed by citing three representative continental ancestral groups(East Asia(CHB),Europe(CEU),and Africa(YRI))from HapMap database.Ancestors through the Structure analysis software for individual source composition and population composition,combining with the building of database matching probability calculation;Source of racial judgment of 46 blind test sample.Results:The system sensitivity is 0.125ng.Twenty and six genotypes were detected in chimpanzee and monkeys,respectively.Except in rs10496971,no more products were found in other animals.The system was able to differentiate intercontinental population.But it was unable to distinguish between East Asian and Southeast Asian,and differentiate between Southern China Han and Hui,Miao,Yi and Tibet.Blind test samples achieved 100%accuracy for intercontinental population source inference.Conclusion 27-plex SNPs multiplex system,with high sensitivity and species specificity,can correctly differentiate the ancestry origins of individuals from African,European and East Asian,respectively,which could provide important clue for criminal case investigating.However,the system has its limitation for distinguishing sub population groups.The more spcific ancestry-informative markers are needed for screening and confirming,to meet the needs of recognizing Southeast Asian and Chinese ethnic populations.
