Objective: To investigate the correlation of mitochondrial morphological structure and function,and to explore the influence of Drp1 and marf on fruit flies indirect flight muscle,Through Drp1(dynamin-related protein1),Marf(mitoechondrial assembly regulatory factor)gene regulation of mitochondrial morphology change.Methods :.Use the MHC-Gal4 promoter,and classic Gal4 / UAS system,gene Drp1 and Marf specifically expressed in fruit flies indirect flight muscle tissue,through gene intervention of knocking out and over expression form to regulate the expression of drosophila indirect flight muscle mitochondria,change the configuration of the mitochondria,observe the abnormal rate of wings and flying ability,and to observe whether the arrangement of the indirect flight muscles and the morphology of mitochondria have changed by electron microscope;by measuring the respiratory function of mitochondria oxidative ATP expression,and further using the Western Blot protein immunoblot technology to detect the the quantity of protein of mitochondrial complex I subunit NDUFS3,and evaluate mitochondrial function control affected by the expression of Drp1 and Marf in drosophila indirect flight muscle.Results: Compared with the control group,gene intervention of knocking out Drp1 and over expression Marf barriers in the drosophila indirect flight muscle,much as for the ability of flying,the rate of abnormal wings is increased;Transmission Electron Microscopy(TEM)can be observed that Drp1,Marf transgenic flies the chest muscle mitochondria present as fusion state,structure is damaged,the number of mitochondria is decreased,drosophila indirect flight muscles arrangement is irregular,muscle fiber shows fracture.The amount of ATP generated by mitochondrial oxidative respiration was reduced,and Western Blot protein detection of mitochondrial oxidative respiration compound I subunits NDUFS3 protein expression levels were significantly lower.Conclusions: Knocking out Drp1 and over expression Marf gene in flies model can change the morphology of mitochondria,influence fruit flies flying function;drosophila model of knocking out Drp1 and over expression Marf gene can make the mitochondrial dysfunction;we speculation whether mitochondrial division limited or fusion,will result in disorder of mitochondrial function.
