Objective:To investigate the clinical significance of two biomarkers HMGB1(High mobility group box-1)and MMP-9(Matrix Met allo-Pro-teinase-9)in type I and II endometrial carcinoma.We measured the expression of HMGB1 and MMP-9 by immunohistochemistry,to explore their relationship with pathologic stage,lymphatic metastasis,and the size of tumors.The experiment aims to provide more evidence for mechanism of endometrial carcinoma,and to enrich the current knowledge of timely diagnosis and treatment.Methods:1.100 cases of type I and II endometrial carcinoma were randomly selected in the study based on clinicopathology characteristics between June 2013 and August2013 at the Maternal and Child Health Hospital of Jiang Xi province.The expression of HMGB1 and MMP9 were analyzed immunohistochemically.The relation of HMGB1 and MMP9 in different clinicopathological types was investigated.2.Paraffin sections were prepared for Hematoxylin and Eosin staining by mounting on superfrost slides.Immunohistochemical staining using the standard avidin–biotin–peroxidase complex(ABC)technique,to localize the two biomarkers source,and in situ zymography to detect activity.3.Staistical analysis: The results were analyzed with Fisher exact test using SPSS 18.0;The correlation of the two biomarkers was also analyzed using Spearman rank correlation analysis.P<0.05 is considered as statistically significance.Results:1.The expressions of HMGB1 and MMP-9 were positive in type I and II endometrial carcinoma compared with normal tissues.The positive expression rates of MMP-9and HMGB1 in type II endometrial carcinoma were both higher than that of type I endometrial carcinoma,the difference was statistically significant(P<0.05).2.The expressions of MMP-9 and HMGB1 in endometrial carcinoma were associated with histological grade,clinical stage,depth of stromal infiltration,vascular invasion,and lymph node metastasis.The differences were statistically significant(P<0.05).3.The expressions of MMP-9 and HMGB1 were positive correlated,the correlation coefficient is 0.721(P<0.05).Conclusion:1.The expressions of MMP-9 and HMGB1 in endometrial carcinoma tissues demonstrate that MMP-9 and HMGB1 plays a critical role in the course of invasion and metastasis of endometrial carcinoma.2.MMP-9 and HMGB1's joint detection indicates that the two biomarkers could be used to early detect and diagnose endometrial carcinoma,as well as to monitor the prognosis of tumor malignancy and to increase patients' survival rate.3.We find that there is positive correlation between MMP-9 and HMGB1,which indicates the two biomarkers are in the same signal pathway.MMP-9 and HMGB1 both play a role in the formation of endometrial carcinoma.The findings provide clinical evidence for the diagnosis and treatment for endometrial carcinoma,more attention should be paid on early detection biomarkers.
