Objective:Explore the defects of repairing rabbit model of full-thickness skin Metod by the application of polycaprolactone-gelatin-lanthanum?PCL-GE-LaCl3?electrospinning nanofibers membrane.Method:Results:The electrostatic spinning technique was used to producte three kinds of membrane which is containing different lanthanum Wt % : PCL-GE?PCL-GE-LaCl3?0.4%??PCL-GE-LaCl3?0.8%?.27 adult rabbit?male and female unlimited?used for the building of the wound..To made six full-thickness skin defects wounds which size is 2.50 x 2.5 cm on both sides on each back of rabbit.Then divided the six wounds into three groups:Vaseline control gurop,PCL-GE group,PCL-GE-LaCl3 group.Each group respectively covers:Vaseline,PCL-GE-nanofibers,PCL-GE-LaCl3?0.4%?-nanofibers,PCL-GE-LaC l3?0.8%?-nanofibers.After 3 days,7 days,15 days each time point,The number of Vaseline control gurop,PCL-GE group,PCL-GE-LaCl3 group respectively :6?6?8?8.Each time point observation the wound,Calculating rate of wound closure,Take the local tissue pathological slices,HE staining,Statistical analysis.Results:?1?PCL-GE,PCL-GE-LaCl3?0.4%?,PCL-GE-LaCl3?0.8%?gurops nanofibers,Fibers are more continuous,uniform,and two kinds of fiber distribution structure,there are more pores.?2?PCL-GE membrane decomposition peak temperature greater than 475 ?,PCL-GE-LaCl3?0.4%?decomposition peak corresponding temperature is 475 ?,PCL-GE-LaCl3?0.8%?decomposition peak corresponding to the temperature of 400 ?,visible,with the increase of lanthanum ion content in composite nanofibers membrane of the initial decomposition temperature drop,showed that lanthanum ion in PCL-GE reduces the thermal stability of the composite nanofibers,easy to decompose.?3?Intraoperative observed electrospinning film are closely with wound paste,translucent shape.After 3 day,postoperatively,electrospinning film still closely with wound paste,surface drying,vaseline group of a small amount of effusion is still visible.After 7 day,containing lanthanum nanofiber membrane surface is clean,membrane under granulation.after 15 day,postoperative wound base finish epithelium,surface material residue.?4?After 3 days,PCL-GE-LaCl3?0.4%?group,the rate of wound closure and vaseline is significantly higher than the control group.PCL-GE-LaCl3?0.8%?group,the closure rate is significantly higher than vaseline control group,and PCL-GE group.After 7 days and 15 days,compared with vaseline control group,there is no difference between the other groups.Conclusion:?1?PCL-GE-LaCl3 nanofibers film with three dimensional form,higher porosity.?2?Along with the increase in lanthanum ion,PCL-GE-LaCl3 electrospinning membrane decreased,the thermal stability of the easy to decompose.?3?PCL-GE-LaCl3 nanometer fiber membrane with good hydrophilicity,biological histocompatibility.?4?Containing lanthanum electrospinning film can reduce wound secretion,may have a local anti-inflammatory effects.?5?Early in the process of wound closure,PCL-GE-LaCl3?0.8%?and PCL-GE-LaCl3?0.4%?of electrospinning film may have a function to promote wound contraction,obvious on PCL-GE-LaCl3?0.4%?group.
