Objective:To investigate the expression of miR-320 a in colon cancer cells and its effects on colon cancer biological process,and preliminarily discuss whether FOXQ1 is a direct target of miR-320 a in HCT116 cells.Methods:1 ? The HCT116 colon cancer cell line was tansfected with miR-320 a mimics,while normal colon cells without being transfected were selected as a control group.The different level of miR-320 a between HCT116 colon cancer cells and normal colon cells was decteted by real-time PCR after transfection.···2?The CCK8 and colony formation assay were used to evaluate the proliferation of two groups of cells(HCT116-mimics and HCT116-NC)after transfection.Transwell chamber was utilized to analyze the effects of miR-320 a on invasion of HCT116 cells.3?Dual luciferase reporter experiments were employed to examine the regulation of miR-320 a on FOXQ1.The mRNA and protein expressions of FOXQ1 was detected by Real-time PCR and Western blot both before and after transfection in colon cancer cells.Results:1?The result of Real-time PCR analysis indicated that miR-320 a mimics significantly increased miR-320 a levels in HCT-116 cells.2?The CCK8 and colony formation assay results showed overexpression of miR-320 a led to inhibit HCT-116 cell proliferation and colony formation.Up-regulation of miR-320 a impaired the invasive abilities of HCT-116 cells.3?The Real-time PCR analysis and Western blot results showed miR-320 a reduced FOXQ1 both on mRNA and protein level,and double-luciferase reporter experiments further confirmed that miR-320 a could directly regulate FOXQ1 expression in colon cancer cells.Conclusion:1?miR-320 a can inhibit cell proliferation and invasion.2?FOXQ1 is a target of miR-320a.
