Objective:The castration resistance and distant metastasis of prostate cancer are difficult to treat,but the mechanism of castration resistance and distant metastasis develop is not clear.A large number of studies have found that androgen receptor splice variants(AR-Vs)may be related to the development and metastasis of prostate cancer.The aim of this study was to explore the clinical significance of AR-V7 in prostate cancer development and bone metastasis.Methods:51 cases prostate tissue specimens were obtained at the First Affiliated Hospital of Nanchang University from 2015.1-2017.4.20 patients with benign prostatic hyperplasia were aged 58-77(average 65.8),20 patients with localized prostate cancer,aged 59-76(average 67.5),11 patients with prostate cancer with bone metastases,aged 52-81(average 66).The age of the three groups did not differ statistically.All patients were diagnosed by pathological examination after surgery.Immunohistochemistry and qRT-PCR were used to detect the expression of AR-V7 protein and AR-V7 mRNA in three different groups.SPSS20.0 statistical software was used to analyze the expression of AR-V7 in three groups of samples.Results:1、The positive expression of AR-V7 protein was in the nucleus or cytoplasm appears brown particles.Immunohistochemistry showed that the positive expression of AR-V7 in three different prostate specimens was: 0 case was positive in benign prostatic hyperplasia specimens;3 cases were positive in localized prostate cancer specimens,the positive rate was 15%.6 cases were positive in the group of prostate cancer with bone metastases specimens,the positive rate was 54.5%.The difference was statistically significant(P <0.005).2、The expression of AR-V7 mRNA in the prostate specimens of patients with benign prostatic hyperplasia,localized prostate cancer and prostate cancer was significantly increased(P <0.05).Conclusion:The expression of AR-V7 in prostate hyperplasia,localized prostate cancer,prostate cancer with bone metastases were increased.AR-V7 may be associated with prostate cancer castration resistance and bone metastasis.
