Objective:To investigate the effects of early exercise training on synaptogenesis after cerebral ischemia-reperfusion injury,and to explore the role of Agrin in the process of exercise promotes synaptogenesis after cerebral ischemia and its underlying mechanisms.Methods:Stroke was induced in adult male Sprague-Dawley rats by left middle cerebral artery occlusion for 60 min,and the animals were then randomly assigned to early exercise group or non-exercise group.Non-exercise group and sham group did not carry out motor training,the rats in early exercise group received a training on a treadmill with gradually increased intensity beginning at 24 h after ischemia,30min per day for a maximum of 14 days.At the corresponding time after cerebral ischemia-reperfusion injury,neurological assessment was performed using neurological deficits scores,the level of anxiety and spontaneous exercise were evaluated by open field and elevated plus maze.In addition,cerebral infarct volume were assessed by TTC and CV staining;Western blot and immunofluorescence staining were used to explore the effect of early exercise training on the time window and cell localization of Agrin expression.And the relationship between Agrin expression and synaptogenesis was analyzed.Results:(1)Compared to non-exercise group animals,animals received early exercise exhibited significantly improved neurological scores and the anxiety level after operation was significantly lower,but not in the recovery of spontaneous activity ability.(2)In consistent with the early improvement of functional outcomes,animals with early exercise had significantly reduced infarct volume,brain water content compared to non-exercise animals.(3)Western Blot and Immunofluorescence results showed that early exercise training significantly promoted synaptogenesis-associated protein expression,upregulated neurotrophic factor levels,promoted synaptogenesis.(4)Early exercise training significantly increased the expression of Agrin;this results suggest that Agrin may be an important protein for motor training to promote synaptogenesis.Conclusion:1?Early exercise training can reduce the infarct volume and promote the recovery of neurological function after stroke?2?Early exercise training significantly promote synaptogenesis-associated protein expression after stroke?3?Early exercise training significantly upregulate the expression of synaptogenesis protein Agrin after stroke?...
