Objective To analyze the differences of serum miR-122 expression in the normal,hepatic fibrosis patients,liver cirrhosis and hepatocellular carcinoma patients.To examine the relation among the level of miR-122,serum alanine aminotransferase and aspartate aminotransferase.To study the possibility of serum miR-122 as a serological marker in diagnosis of liver fibrosis.Furthemore to provide guidance for the clinical evaluation of liver disease prognosis,early diagnosis of liver cirrhosis and hepatic fibrosis.Methods Fifteen people in the control group and fourty five patients in the experimental group including fifteen patients with hepatic fibrosis,fifteen patients with liver cirrhosis,and fifteen patients with hepatocellular carcinoma,were enrolled in this study.Thity-two patients with positive hepatitis B surface antigen in the control and experimental groups were also enrolled.The level of serum miR-122 was examined by fluorescence quantitative PCR in all of them.Alanine aminotransferase and aspartate aminotransferase were detected in the serum of the control group and the experimental group.To analyze the differences of serum miR-122 expression in different groups,and to examine the relation among the level of miR-122,serum alanine aminotransferase and aspartate aminotransferase.Results Univariate analysis of variance showed that the expression of miR-122 in the four groups of patients with normal,liver fibrosis,liver cirrhosis and liver cancer was not all the same.The difference was statistically significant(F=34.843,P<0.05).The level of serum miR122 in liver fibrosis group,liver cirrhosis group and hepatocellular carcinoma group were higher than those in control group.The expression level of miR-122 in liver fibrosis group was the highest,and there was no difference between liver cirrhosis group and hepatocellular carcinoma group.The expression of serum miR-122 in the experimental group and the positive group of hepatitis B surface antigen was also higher than that in the control group.The correlation coefficients of serum miR-122 with alanine aminotransferase and aspartate aminotransferase were 0.568 and0.514 respectively.The area under the ROC curve of serum miR-122 in diagnosis of hepatic fibrosis was 0.843±0.064.Conclusions1.The level of serum miR-122 in patients with liver fibrosis,cirrhosis,hepatocellular carcinoma and positive hepatitis B surface antigen were higher than those in normal subjects.2.Serum miR-122 detected by fluorescence quantitative PCR can reflect the degree of liver injury.3.Serum miR-122 detected by fluorescence quantitative PCR is supposed as a potential serological marker for the diagnosis of hepatic fibrosis.
