The tissue and serum MUC16(CA125)levels and its correlation with IL-6 and IL-8 in adenomyosisAbstract:Objective: The aim of the study is to detect the difference between the patients with adenomyosis and the normal control group including the main clinical symptoms(dysmenorrhea,menorrhagia),signs(uterine volume)and serological indexs and to investigate the expression of MUC16 ? IL-6 ? IL-8 m RNA in eutopic/ectopic endometrium and the level of serum CA125 ? IL-6 ? IL-8 in adenomyosis,which is associated with the local inflammatory pathology.Contents: We tested the gene expression of MUC16(CA125),IL-6,IL-8 in the eutopic endometrium with adenomyosis(Eu E),the ectopic endometrium with adenomyosis(EE)and the endometrium without adenomyosis(CE),and serum of CA125,IL-6,IL-8 levels in a series of 31 petients with adenomyosis.Methods:1.Pearson method was used to analyze the correlation between the 31 cases of adenomyosis and 24 cases of normal control group including the main clinical symptoms(dysmenorrhea,menorrhagia),characteristic(uterine volume)and serological indexes.2.The gene expression of MUC16(CA125)in the eutopic endometrium with adenomyosis(Eu E),the ectopic endometrium with adenomyosis(EE)and the endometrium without adenomyosis(CE)was tested by RT-PCR in a series of 31 petients with adenomyosis;The gene expression of IL-6 in the eutopic endometrium with adenomyosis(Eu E),the ectopic endometrium with adenomyosis(EE)and the endometrium without adenomyosis(CE)was tested by RT-PCR in a series of 31 petients with adenomyosis;The gene expression of IL-8 in the eutopic endometrium with adenomyosis(Eu E),the ectopic endometrium with adenomyosis(EE)and the endometrium without adenomyosis(CE)was tested by RT-PCR in a series of 31 petients with adenomyosis;3.Serum of CA125 levels were determined by chemiluminescence immunoassay in a series of 31 petients with adenomyosis;serum of IL-6 levels were determined by chemiluminescence immunoassay in a series of 31 petients with adenomyosis;serum of IL-8 levels were determined by chemiluminescence immunoassay in a series of 31 petients with adenomyosis;4.The Pearson method was used to analyze correlation between the expression of MUC16 m RNA and the expression of IL-6 and IL-8 m RNA in normal endometrium,eutopic endometrial tissue and ectopic endometrial tissue.5.The Pearson method was used to analyze the correlation between the expression of MUC16 m RNA and the levels of serum CA125,IL-6 and IL-8 in normal endometrium;The Pearson method was used to analyze the expression of MUC16 m RNA eutopic endometrium with adenomyosis and the levels of serum CA125,IL-6 and IL-8;The correlation between serum CA125,IL-6 and IL-8 levels in ectopic endometrium with adenomyosis was analyzed by Pearson method;6.The Pearson method was used to analyze the correlation between the levels of serum CA125 in patients with adenomyosis and IL-6,IL-8 m RNA expression in eutopic endometrium and ectopic endometrium,and serum levels of IL-6 and IL-8;The correlation between the levels of serum CA125 in control and IL-6,IL-8 m RNA expression in control endometrium,and serum levels of IL-6 and IL-8 were analyzed by the Pearson method.Results1.Clinical features: menstruation in patients with adenomyosis was improved significantly compared with the normal control group(P<0.01);imaging examination showed that there was significant difference between the patients with adenomyosis and normal control group(P<0.01);Serum hemoglobin(Hb)with adenomyosis was lower than the normal control group(P<0.01);2.The gene expression of MUC16 was significantly higher in Eu E and EE than that in CE(P<0.01);The gene expression of IL-6 was significantly higher in Eu E and EE than that in CE(P<0.01);The gene expression of IL-8 was significantly higher in Eu E and EE than that in CE(P<0.01);3.Serum CA125 level was significantly higher in cases of adenomyosis when compared with those without adenomyosis(p < 0.01);Serum IL-6 level was significantly higher in cases of adenomyosis when compared with those without adenomyosis(p < 0.01);Serum IL-8 level was significantly higher in cases of adenomyosis when compared with those without adenomyosis(p<0.01).4.A positive correlation between the gene expression of MUC16 was found in patients with and without adenomyosis(p<0.01).A positive correlation between the gene expression of IL-6 was found in patients with and without adenomyosis(p<0.01).A positive correlation between the gene expression of IL-8 was found in patients with and without adenomyosis(p<0.01).5.There was no correlation between gene expression of MUC16 in control endometrium and serum CA125,IL-6,IL-8 levels in control groups(p>0.05);and there was no correlation between gene expression of MUC16 in eutopic endometrium and serum CA125,IL-6,IL-8 levels in patients with adenomyosis(p>0.05).Also,there was no correlation between gene expression of MUC16 in ectopic endometrium and serum CA125,IL-6,IL-8 levels in patients with adenomyosis(p>0.05).6.There was no correlation between serum CA125 and IL-6,IL-8 in control both and serum and gene expression levels in control endometrium(p>0.05).And there was no correlation between serum CA125 in patients with adenomyosis and IL-6,IL-8 both serum and gene expression levels in eutopic endometrium(p>0.05).Also,there was no correlation between serum CA125 in patients with adenomyosis and IL-6,IL-8 expression both serum and gene expression levels in ectopic endometrium(p>0.05).Conclusions: The results showed that the expression of MUC16 m RNA in the adenomatous lesion significantly increased and correlated with the expression of local inflammatory factor IL6 / IL8.The serum CA125 concentration could not reflect the expression of MUC16 m RNA in the tissue of the adenomyosis and was not correlated with local inflammation and systemic inflammatory stress.The value and potential significance of MUC16 m RNA expression and serum CA125 levels in patients with adenomyosis need to be further studied.
