| Objective: To observe the influence of different storage time erythrocyte on the pathologic changes of lung tissue,surfactant protein A(SP-A),oxidative stress associated protein,cell factor,endotoxin shock rabbit were infused with different storage time erythrocyte.In order to explore the effect of different storage time erythrocyte on lung injury in endotoxin shock rabbits,and comprehensive evaluate the safety and efficacy of different storage time red blood cell in endotoxin shock model.Methods: Select 74 clean grade healthy male New Zealand rabbits,weighing 2.0-2.5kg,10 of them using the random number table divided into blood donation group other 64 animails were divided into 8 groups(n=8 each): group C(black control group),group S(loss blood group),group L(endotoxin group),group SL(loss blood and endotoxin),group R(red blood cell group R0-R3,four subgroup).Rabbits in group S,group SL,group R were weighed on the day before the experiment,15% blood volume were collected from the ear central artery of each group of rabbits(blood volume was calculated by weight*7%).At the beginning of the experiment 3% 1.5ml/kg sodium pentobarbital was injected into the ear vein.Rabbits in each group were punctured internal carotid artery and placed into the catheter.Group L,SL and R were infused with Lipopolysaccharides(LPS)5mg/kg(dissolved in 1ml saline)by the ear vein,group C and S were injected with the same amount of normal saline.When the mean arterial pressure(MAP)decreased more than 25% and oxygenation index(OI)decreased to 300 mm Hg,the model was successfully established.After intravenous injection of LPS 2 hours,each group of group R was infused with 5% blood volume of storage erythrocyte,respectively,with one day storage of red blood cells(group R0),one week storage of red blood cells(group R1),two weeks of storage of red blood cells(group R2),three weeks of storage of red blood cells(group R3),and appropriate amout of lactated Ringer’s solution to maintain MAP>65 mm Hg.Group C,group S,group L and group SL were also given appropriate amout of lactated Ringer’s solution to maintain MAP>65 mm Hg.In addition,serum Interleukin-1β(IL-1β),Interleukin-8(IL-8),Tumor Necrosis Factor-α(TNF-α)were detected at three point-before the intravenous injection of LPS(T1),injection of LPS 2h(T2),injection of LPS 6h(T3).The rabbits were killed at T3 time,and then testing the pathological examination of inferior lobe of left lung,the Wet/dry Ratio(W/D)of inferior lobe of right lung,detecting the changes of Myeloperoxidase(MPO),Malondialdehyde(MDA),Superoxide Dismutase(SOD),Nitric Oxide Synthase(NOS)after the superior lobe of right lung homogenate.Results:(1)Compared with group C and S,the pathological score of lung tissue of group L,SL,R0,R1,R2,R3 incressed(P < 0.05),compared with group L,SL,R0 the pathological score of lung tissue of group R2 and R3 incressed(P < 0.05)(2)At time T2 and T3,compared with group C and S,the ratio to W/D in group L,SL,R0,R1,R2,R3 were significantly increased(P < 0.05),compared with group L and SL,the ratio to W/D in group R3 were significantly increased(P < 0.05);(3)Compared with group C and S,the activity of MPO and the expression of MDA were significantly increased in group L,group SL,group R0,group R1,group R2,group R3(P < 0.05)but the activity of SOD were significantly decreased;Compared with group L and SL,the activity of MPO were significantly increased and the activity of SOD were significantly decreased in group R1,R2,R3(P < 0.05),the expression of MDA were significantly increased in group R0,R1,R2,R3(P < 0.05).(4)Compared with group C and S,the activity of NOS were significantly incressed in group L,SL,R0,R1,R2,R3(P < 0.05)(5)Compared with group Cand S,the expression of SP-A were significantly decressed in group L,SL,R0,R1,R2,R3(P < 0.05),Compared with group L and SL,the expression of SP-A were significantly decressed in group R2,R3(P < 0.05)(6)At time T2 and T3,Compared with group C and S,TNF-alpha and IL-1 beta and IL-8 had significantly increased in group L,SL,R0,R1,R2,R3(P < 0.05),at time T3,Compared with group L,R0 and R1,IL-8 were increased in group R3(P < 0.05);IL-1 beta in group R3 were increased compared with other groups(P < 0.05).Conclusion:(1).Oxidative stress is involved in lung injury induced by endotoxin shock.(2).With the prolongation of erythrocyte storage time,MDA and MPO increased and SOD decreased,but did not affect the formation of NOS and SP-A in endotoxin shock rabbits.(3).The oxidized derivatives of storage erythrocytes will aggravate the lung injury induced by endotoxin shock.It’s better to choose red blood cells within 14 days when the red blood cells are needed. |
