ObjectiveIn this vitro study,with the new angle of view of inflammation--insulin signal transduction pathway--IR(Insulin Resistance),we aimed to investigate the effect and mechanism of the total saponins of momordica charantia on cellular glucose intake and the expression and the secretion of TNF-? and IL-6 in insulin resistance 3T3-L1 adipocytes so as to reveal the mechanism of total saponins of momordica charantia in improving insulin resistance by inflammatory factors.Methods1 To determine the proliferative effect of the total saponins of momordica charantia on the normal 3T3-L1 adipocytes and screen of drug concentration by MTT assay.2 Pre-adipocyte was induced into mature adipocyte step by step with the induced liquid,and the formation of the adipocyte was identified by using oil red“O” staining method.3 The insulin resistant cell model was established with Dexamethasone.4 The content of glucose in the observation group cultures of change was detected by using glucose oxidase method after adding total saponins of momordica charantia for 24 h.5 The m RNA expression of TNF-? and IL-6 in adipocyte was detected byusing Realtime-Polymerase Chain Reaction(RT-q PCR).6 The secretion of TNF-? and IL-6 was detected by ELISA.Results1 The preferable reaction time for culture with 3T3-L1 cells of total saponins of momordica charantia was 24 h,while the concentrations were momordica charantia high concentration group(5mg/m L)and momordica charantia low concentration group(2.5mg/m L).2 The fat drips were stained by oil red O after induction of differentiation by12 days.3 The insulin resistance model was duplicated after dealing with dexamethasone for 48h(P<0.01).4 Compared to the model group,when the IR 3T3-L1 fat cells were intervened with total saponins of MC,it was found that glucose intake increased significantly(P<0.01).5 Compared to the model group,after the IR 3T3-L1 fat cells were dealed with total saponins of MC,the expression TNF-? and IL-6 had decreased significantly(P<0.01)as well as the protein secretion of TNF-? and IL-6 had decreased significantly(P<0.01).Conclusions1 Total saponins of momordica charantia could significantly improve the glucose intaking of insulin resistance 3T3-L1 fat cells,as well as the normal3T3-L1 fat cells had no obvious effect on the sugar content.2 Total saponins of momordica charantia could obviously decrease insulin resistance 3T3-L1 fat cells abnormal TNF-?,IL-6m RNA levels and the levels of protein secretion.
