Objective This study was designed to make the mouse model of sarcopenia by dexamethasone,and to explore the effect of resistance training and creatine supplementation in a mouse model of sarcopenia.Methods C57 BL / 6 male mice were randomly divided into two groups: model group(group D)32,control group(N group)8.Group D was injected subcutaneously into dexamethasone.Group N was injected subcutaneously with 0.9% saline for 19 consecutive days to establish a mouse model of sarcopenia.The muscle mass and muscle strength of mice were detected by Echo MRITM and Grip Strength Meter.This model was evaluated by the change of muscle mass and muscle strength in mice.The model mice were randomly divided into four groups: resistance training combined with creatine supplement(group A),resistance training(group B),creatine supplement(group C)and control(Group D),8 in each group.Resistance training 5d / w,creatine intragastric 7d / w.The muscle mass was measured by Echo MRI TM and the muscle strength by Grip Strength Meter.Muscle function was detected by wheel fatigue and water maze.The effect of the intervention experiment was evaluated bychanges in muscle mass,muscle strength and muscle function.Results A lower of muscle mass and muscle strength were present in sarcopenia model mice than in mice of controls(P <0.05).After intervention,the body weight decreased in resistance training combined with creatine supplement group compared with resistance training group,creatine supplement group and control group,P <0.05.The muscle mass and muscle strength were significantly higher in resistance training combined with creatine supplement group than resistance training group,creatine supplement group and control group(P <0.05).The number of drops in resistance training combined with creatine supplement group was significantly lower than resistance training group,creatine supplement group and control group(P <0.05).The swimming speed of water maze test was significantly higher than that of resistance training group,creatine supplement group and control group(P <0.05).Conclusions Resistance training combined with creatine supplement can increase muscle mass and enhance muscle strength and function in a mouse model of sarcopenia.Resistance training combined with creatine supplementation is an effective method of interfering with sarcopenia in mice.
