ObjectTo investigate the influence of hydrogen sulfide upon apoptosis in adipose tissue of diabetic mice and its mechanism.MethodsHealthy male C57BL/6J mice were divided into 5 groups;Negative control group,Diabetes group,Low doze treatment group,High doze treatment group,and Positive control group.Caspase3 activity,Bax,Bcl2,and PPAR-? protein as well as m RNA expression levels were measured after 8 weeks of treatment.3T3L1 cell line was induced to adipocytes,and divided into 8 groups,Low glucose group,Low glucose treatment group,Low glucose PPAR-? blocked group,Low glucose PPAR-? blocked treatment group,High glucose group,High glucose treatment group,High glucose PPAR-? blocked group and High glucose PPAR-? blocked treatment group.Caspase3 activity,Bax,and Bcl2 protein expression levels were measured after cells were harvested.ResultsAnimal experiments results were showed as followings.Compared to negative control group,Caspase3 activity and Bax expression levels increased while Bcl2 and PPAR-? expression levels decreased in diabetes group.Compared to diabetes group,Caspase3 activity and Bax expression levels decreased while Bcl2 and PPAR-? expression levels increased in low doze treatment group,high doze treatment group and positive control group.Cell experiments results were showed as followings.Compared to low glucosegroup,Caspase3 activity and Bax expression levels increased while Bcl2 level decreased in high glucose group.Compared to high glucose group,Caspase3 activity and Bax expression levels decreased while Bcl2 level increased in high glucose treatment group.Compared to high glucose treatment group,Caspase3 activity and Bax expression levels increased while Bcl2 level decreased in high glucose PPAR-? blocked treatment group.There is a antagonism effect between high glucose and hydrogen sulfide as well as hydrogen sulfide treatment and PPAR-? blocked treatment,while synergistic effect between PPAR-? blocked treatment and high glucose.ConclusionHydrogen sulfide inhibited apoptosis of adipose tissue of diabetic mice may account for up-regulating of PPAR-?.
