| Objective: To investigate the relationship and correlation between miRNA-21 and pulmonary arterial hypertension(PAH)by comparing the expression level of microRNA-21 with the commonly used index measured by cardiac catheterization.To explore the value in assessing the severity of PAH.Methods: Select patients in cardiovascular department of the Guangxi Zhuang Autonomous Region People's Hospital from November 2016 to March 2017.Measure blood pressure and blood gas analysis of atrial ventricular cavity by cardiac catheterization.61 cases were collected,including 20 males and 41 females.According to the latest diagnostic criteria of pulmonary hypertension published by the European Society of Cardiology in 2015,the patients were divided into two groups: 17 patients with elevated pulmonary artery pressure(Group A)and 44 patients with normal pulmonary artery pressure(group B).Extract 5ml venous blood to detect miRNA-21.Total RNA was extracted by Trizol method.Finally,total RNA was reverse transcribed into cDNA,and realtime quantitative PCR was performed.The expression of miRNA-21 was calculated according to the 2-? ? CT method.Results:(1)MPAP of patients in Group A and B is(41.24±13.06)mmHg?(19.90±2.41)mmHg(P<0.05),PVR of patients in Group A and B is(4.97±0.91)Wood ?(3.40±0.54)Wood(P<0.05),PVRi of patients in Group A and B is(7.74±1.18)?(5.28±1.03)(P<0.05),Qp/Qs of patients in Group A and B is(1.58±0.42)?(1.31±0.21)(P<0.05);(2)The level of miRNA-21 of patients in Group A and B is(5.26±1.06)?(3.69±0.86)(P<0.05);(3)Pearson correlation coefficient between miRNA-21 and mPAP is 0.631(P<0.05),pearson correlation coefficient between miRNA-21 and PVR is 0.522(P<0.05),pearson correlation coefficient between miRNA-21 and PVRi is 0.503(P<0.05),there is no correlation between miRNA-21 and Qp/Qs(P=0.165).Conclusion: The expression level of miRNA-21 in patients with PAH is increased significantly,miRNA-21 can be used as a potential indicator in pulmonary arterial hypertension detection and assessment. |
