Objectives : To compare the effects of different concentrations of sevoflurane on hyperoxia-induced lung injury in rats.Methods:1.60 rats were divided into blank control group(group C,n=10)and experimental group(group S,n=50).group S used 5L / min or so oxygen to replicate hyperoxia lung injury model,in which the oxygen concentration to maintain more than 95%,exposure time of 48 hours,replication of hyperoxia-induced injury animal model;2.SPSS17.0 random number generator was used to divide 50 rats in group E into 5 groups,which were inhaled for 0%,1%,1.5%,2%,and sevoflurane for 1 hours,and were named as group S0,group S1.0,group S1.5,group S2.0,group S2.5 respectively;3.At 48 hours after completion of hyperoxia treatment,and sevoflurane was inhaled for 1 hour at two time points,By 10% of the chloral hydrate anesthesia by abdominal aorta extraction of arterial blood,serum and collection of left and right lung tissue;4.Arterial blood gas analysis PO2,PCO2.Serum TNF-?,IL-8 and IL-6 concentrations were measured by ELISA.The wet/dry(W/D)ratio of the left lobe of the lung tissue was measured.The pathological changes of the right lung tissue were observed by HE staining.Score reference indicators: alveolar cavity congestion,bleeding,alveolar or vascular wall neutrophil infiltration aggregation,alveolar wall thickening and transparent film formation.Results:1.Compared with group C,the PO2 of group S decreased,PCO2 increased,TNF-?,IL-8 and IL-6 increased,W/D ratio increased,pathological damage score increased,significant difference(P <0.05),There was no significant difference between the S0 to S2.5 groups after 48 hours of hyperoxia treatment(T1)(P> 0.05).2.After 1 hour of treatment with sevoflurane(T2):(1).Compared with group C,PO2 decreased,PCO2 increased,TNF-?,IL-8 and IL-6 increased,W/D ratio increased,pathological damage score increased in group S,significant difference(P <0.05);(2).Comparison with seven halothane before treatment,PO2 increased,PCO2 decreased,TNF-?,IL-8 and IL-6 decreased,W/D ratio decreased,pathological damage score decreased in group S1.0 to S2.5,significant differencethe(P <0.05).but there was no significant change in group S0(P> 0.05);(3).Compared with S2.0 group,PO2 decreased,PCO2 increased,TNF-?,IL-8 and IL-6 increased,W/D ratio increased,pathological damage score increased in the group S1.0 and S1.5,significant difference(P <0.05);(4)Compared with S2.0 group,there were no significant differences in PO2,PCO2,TNF-?,IL-8 and IL-6 concentrations,W/D ratio and pathological score in S2.5 group(P> 0.05).Conclusion:1.The rat models of hyperoxia induced lung injury was induced by 95% concentration of oxygen for 48 hours;2.Sevoflurane can effectively reduce the degree of lung injury caused by hyperoxia in rats;3.2% concentration of sevoflurane has a good protective effect on hyperoxia-induced lung injury in rats.
